Therapeutic effect of human umbilical cord mesenchymal stem cells on endometritis in mice
Objective To investigate the effect of human umbilical cord mesenchymal stem cells(hUC-MSCs)in repairing the mice model of endometritis in vivo.Methods Lipopolysaccharide(LPS)was used to induce acute inflammation in mice model of endometritis,grouping:control group(PBS),model group(LPS),early treatment group(LPS+hUC-MSCs,6 h),and advanced treatment group(LPS+hUC-MSCs,12 h).Morphological changes of mouse uterus were directly observed,histological changes of endometrium were observed by HE staining,ELISA method for detecting mouse uterine inflammation index MPO and related immune index TNF-α,IL-6 and IL-1β changes.Results Compared with control group,the model group showed significant inflammation through visual observation and histological examination,and the content of MPO,TNF-α,IL-1β,and IL-6 in uterine tissue significantly increased.hUC-MSCs treatment group showed reduced inflammatory reactions in morphological and histological appearance,and the value of MPO,TNF-α,IL-1β and IL-6 showed a different degree of decline.After the formation of inflammation,early perfusion of hUC-MSCs showed the most significant decrease in inflammatory indicators.Conclusion Direct intrauterine infusion of hUC-MSCs can alleviate LPS induced endometritis.
