Objective To study the effects of rs4454537 polymorphism of Gremlin-2 gene and abnormal level of 25-hydroxyvitamin D3[25(OH)D3]on osteoporosis in postmenopausal women.Methods 160 postmenopausal osteoporosis patients admitted to Xingtai People's Hospital from February 2020 to October 2023 were selected as the study group,and 160 healthy menopausal women who underwent physical examination during the same period were selected as the control group.Bone mineral density(BMD)and 25(OH)D3 levels of the two groups were compared,and the correlation of which was analyzed.TaqMan probe method was used to detect the polymorphism of Gremlin-2 gene rs4454537 in the blood of patients,and the gene distribution,serum 25(OH)D3 level and BMD of patients with different genotypes were compared,and the influencing factors of osteoporosis in postmenopausal women were analyzed.Results The expression level of 25(OH)D3,BMD and T value in the study group were lower than those in the control group(P<0.05).The expression level of 25(OH)D3 was positively correlated with BMD by Pearson analysis(r=0.638,P<0.05).There were three genotypes at the rs4454537 locus of Gremlin-2 gene:TT,TC and CC.The frequencies of T allele and TT genotype in the study group were lower than those in the control group(x2=19.643,P<0.001,x2=18.321,P<0.001).BMD and 25(OH)D3 levels in patients with different genotypes were significantly different,and BMD and 25(OH)D3 levels in patients with TT genotype were significantly higher than those in patients with TC and CC genotype(P<0.05).Logistic regression analysis showed that rs4454537 genotype of Gremlin-2 gene and 25(OH)D3 level were independent influencing factors for postmenopausal osteoporosis(P<0.01).Conclusions The rs4454537 locus polymorphism of Gremlin-2 gene and serum 25(OH)D3 level are closely related to osteoporosis in postmenopausal women,and TT genotype patients have a high level of serum 25(OH)D3 level,which may decrease the risk of osteoporosis.
