目的 探究五加生化胶囊对流产模型大鼠子宫内膜修复作用的机制.方法 随机选取10只雌性大鼠作为空白对照组(空白组),其余60只雌性大鼠与30只雄性大鼠按照2:1的比例合笼饲养,将妊娠大鼠灌胃米非司酮和米索前列醇制备药物流产大鼠模型,造模成功后的40只大鼠随机分为5组,即模型组、低剂量组、中剂量组、高剂量组和阳性对照组,每组8只.检测各组大鼠子宫出血量、凝血指标及血清中雌二醇(estradiol,E2)、孕酮(progesterone,P)、纤维连接蛋白(fibronectin,FN)和层粘连蛋白(laminin,LN)水平;苏木素-伊红(hematoxylin-Eosin,HE)染色检测大鼠子宫组织病理学变化;蛋白质印迹检测子宫组织中基质金属蛋白酶-9(MMP-9)、金属蛋白酶组织抑制因子1(TIMP1)蛋白表达.结果 和空白组相比,模型组大鼠子宫出血量,血浆中PT、TT、APTT,血清中P、FN和LN水平均明显增加,血清中E2水平明显降低(P<0.05);和模型组相比,低、中、高剂量组和阳性对照组大鼠子宫出血量,血浆中PT、TT、APTT,血清中P、FN和LN水平明显减少,血清中E2水平明显升高(P<0.05).模型组大鼠子宫组织中MMP-9蛋白表达高于空白组,TIMP-1蛋白表达低于空白组(P<0.05);低、中、高剂量组和阳性对照组大鼠子宫组织中MMP-9蛋白表达低于模型组,TIMP-1蛋白表达高于模型组(P<0.05).结论 五加生化胶囊可治疗药物流产后大鼠异常子宫出血,其作用机制可能为改善凝血功能、调节雌激素水平,调节子宫组织中MMP-9和TIPM-1平衡,影响FN和LN水平,从而加速受损子宫内膜基质的修复.
Mechanism study on the repair effect of Wujia biochemical capsule on endometrium of abortion model rats
Objective To investigate the mechanism of the repair effect of Wujia biochemical capsule on the endometrium of abortion model rats.Methods Ten female rats were randomly selected as the blank control group(blank group),and the remaining 60 female rats were housed in a 2:1 ratio with 30 male rats in a combined cage.The drug abortion rat model was prepared by gavage of mifepristone and misoprostol in pregnant rats,and the 40 rats successfully modeled were randomly divided into model group,low-dose group,medium-dose group,high-dose group and positive control group,8 rats in each group.Measured the amount of uterine bleeding,coagulation indicators,and the levels of estradiol(E2),progesterone(P),fibronectin(FN),and laminin(LN)in rat serum in each group of rats;hematoxylin eosin(HE)staining was used to detect pathological changes in rat uterine tissue;protein blotting was used to detect the expression of matrix metalloproteinase-9(MMP-9)and tissue inhibitor of metalloproteinase-1(TIMP1)proteins in uterine tissue.Results Compared with the blank group,the uterine bleeding,plasma levels of PT,TT,APTT,serum levels of P,FN and LN were significantly increased and serum levels of E2 were significantly decreased in the model group(P<0.05);compared with the model group,the uterine bleeding,plasma levels of PT,TT,APTT,serum levels of P,FN and LN were significantly decreased in the low,medium and high dose groups and the positive control group,and the serum E2 level was significantly increased(P<0.05).The expression of MMP-9 protein in uterine tissue of rats in the model group was higher than that in the blank group,and the expression of TIMP-1 protein was lower than that in the blank group(P<0.05).The expression of MMP-9 protein in uterine tissue of rats in low,medium and high dose groups and positive control group was lower than that in model group,and the expression of TIMP-1 protein was higher than that in model group(P<0.05).Conclusion Wujia biochemical capsule can treat abnormal uterine bleeding in rats after medical abortion.Its mechanism may be to improve coagulation function,regulate estrogen level,regulate the balance of MMP-9 and TIPM-1 in uterine tissue,affect FN and LN levels,and accelerate the repair of damaged endometrial matrix.
