国产胰酶肠溶片的质量研究与风险分析

研究国产胰酶肠溶片的质量,分析产品目前存在的质量风险和质量标准中需要完善的指标.按照《中华人民共和国药典》2015年版二部的法定方法对全国范围内抽验的49批样品进行检验,并结合探索性质量研究,主要包括溶出度、罩泡密闭性、水分、残留溶剂、效价测定、种属鉴别等,对性状不合格的原因及反映的质量风险进行深入探究.对国产制剂与进口参照制剂进行了效价测定和溶出曲线的一致性评价,论证了将绝对效价测定方法修订为相对效价测定方法的必要性.通过分析认为本品主要在有效性上存在风险隐患,我国药典标准较为简单,应增订溶出度、水分,修订效价测定等影响产品质量的关键检测指标,为标准完善和改进产品质量提供参考依据.
Quality Research and Risk Analysis of Domestic Pancreatin Enteric-coated Tablets
To study the quality of domestic pancreatin enteric-coated tablets,and analyze the existing quality risks of products and the testing items that need to be improved in the quality standard,forty-nine batches of products were included nationwide and investigated.Examination and exploratory studies,including dissolution,bubble tightness,water content,residual solvent,enzyme activity and species identification were carried out according to the Chinese pharmacopoeial method (Volume Ⅱ,2015 Edition).Further investigation was employed for cause and quality risk of description disqualification.Consistency evaluations of enzyme activities and dissolution curves were carried out between domestic and imported similar preparations.Revision was proposed that absolute enzyme activity needed to be replaced by relative enzyme activity.Its necessity was also demonstrated.Results showed that potential risks existed in effectiveness of the products.The Chinese pharmacopoeial standard is relatively simple and key indexes which influence the quality of products need to be improved,including new admission of dissolution and water content,and revision of enzyme activity.The current study provided research ideas and data for perfecting the quality standard and improving product quality.
