Investigation on Concentration Determination and Tissue Distribution of Natamycin in New Zealand White Rabbit Eyes
In this study,a LC-MS/MS analytical method was developed for determination of natamycin(1)to assess its distribution in various ocular tissues of New Zealand white rabbits.The results revealed that,after intravitreal administration of 1 to New Zealand white rabbits,1 was widely distributed in various ocular tissues,particularly in the posterior segment of the eye.The cmax and AUC0→24 of 1 in retina and vitreous were 46 400 and 4 620 ng/g,232 000 and 29 300 h·ng·g-1,respectively.The results demonstrated that the 1 amounts were higher in both retina and vitreous than in other ocular tissues.After 24 hours post-dosing,the drug concentration remained high level in all ocular tissues,indicating a longer retention of 1.Given this high specificity and reliable results,this established LC-MS/MS method is suitable for investigating distribution of 1 in ocular tissues and can be used to explore further potential treatments for ocular diseases with 1.
