原位凝胶是一种以溶液状态给药,在用药部位迅速发生相变,成为半固体或固体凝胶的新型制剂;并且,能对温度、光照、pH值、亲/疏水性、离子强度等外界因素的变化做出响应.原位凝胶具有良好的生物相容性、生物黏附能力以及缓控释特性,在眼部、鼻腔、口服、透皮、注射以及阴道等多种途径中均展现出独特的应用优势.该文结合各种给药途径的特性,对比了传统剂型与原位凝胶剂的优势和缺陷,介绍了原位凝胶剂在多种给药途径中应用的研究进展,并展示了一些已上市的原位凝胶产品,为原位凝胶制剂的研发提供参考.
In situ gel is a novel formulation which is administered in solution and can rapidly occur phase transformation at the site of application to become semi-solid or solid gel.In addition,it can respond to the changes in external factors such as temperature,light,pH value,hydrophily/hydrophobicity,ionic strength.In situ gel has shown unique advan-tages in a variety of applications,including ophthalmic,nasal,oral,transdermal,injectable and vaginal routes,due to its excellent biocompatibility,bioadhesion ability,sustained-and controlled-release properties.In this paper,the advantages and disadvantages of traditional dosage forms and in situ gel are compared according to the characteristics of various routes of administrations.The research progress of in situ gel in above routes is introduced,and some approval in situ gel products are presented.
