LC-MS法测定托格列净中的4-二甲氨基吡啶及其氮氧化物

建立了LC-MS法测定托格列净(1)中的4-二甲氨基吡啶(2)和4-二甲氨基吡啶氮氧化物(3).采用ACQUITY UPLC BEH C18色谱柱(2.1 mm×50 mm,1.7 μm)和电喷雾离子源,多反应监测模式,以甲醇为流动相A,0.1%甲酸水溶液为流动相B,梯度洗脱.结果表明,2和3分别在15.40~616.00 ng/mL和15.58~623.20 ng/mL内线性关系良好,检测限分别为3.08、3.12 ng/mL,2和3的平均回收率(n=9)分别为104.33%和101.54%,RSD分别为3.32%和3.10%.建立的方法灵敏度好、选择性强、准确度高,可用于1中的2和3的测定.
Determination of 4-Dimethylaminopyridine and Its N-Oxide in Tofogliflozin by LC-MS
A LC-MS method was established to determine the 4-dimethylaminopyridine(2)and 4-dimethyl-aminopyridine N-oxide(3)in tofogliflozin(1).The ACQUITY UPLC BEH C18 column(4.6 mm×150 mm,5 pm)was used,with the electrospray ionization and multiple response monitoring mode.The analysis was carried out in the gradient elution mode with methanol as mobile phase A,and 0.1%formic acid aqueous solution as mobile phase B.The results showed that it was linear for 2 and 3 in the ranges of 15.40-616.00 ng/mL and 15.58-623.20 ng/mL,respectively.The limits of detection for 2 and 3 were 3.08 and 3.12 ng/mL,respectively.The average recoveries were 104.33%and 101.54%,with RSDs of 3.32%and 3.10%,respectively.The established method is sensitive,selective and accurate,which can be used for the determination of 2 and 3 in 1.
