Methyltransferase 16(METTL16)is a newly discovered N6-methyladenosine(m6A)methyltransferase,which plays an increasingly prominent role in tumor research and has been confirmed to be abnormally expressed in various tumor diseases, playing varying degrees of regulatory roles.METTL16 can regulate the stability of target RNA by exerting the activity of methyltransferase, thereby exerting tumor-promoting or inhibiting effects in different tumors by influencing cell cycle transition, promoting glycolysis and inhibiting ferroptosis. However, there are also reports that METTL16 can promote translation by interacting with eukaryotic translation initiation factors, and promote the occurrence and development of tumors without relying on its methyltransferase activity.Therefore,METTL16 holds important value in the research of tumor diseases, and it is expected to become a new biomarker for early diagnosis and treatment of tumors. This review focuses on the research progress of METTL16 in the occurrence and development of tumors.
