Exploration of the mechanism of action of Jianpi Wenyang prescription in treating primary premature ejaculation based on network pharmacology
Objective To analyze the potential mechanism of action of Jianpi Wenyang prescription in treating primary premature ejaculation(PPE)based on network pharmacology.Methods The Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform(TCMSP)and the Bioinformatics Analysis Tool for Molecular mechANism of Traditional Chinese Medicine(BATMAN-TCM)platform were used to collect the chemical active ingredients of all drugs in Jianpi Wenyang prescription,including corresponding main targets.The relevant action targets of PPE were collected using Genecards and Drugbank databases.The effective component targets were intersected with the pain targets,the intersection targets associated with the effective component and the pain were obtained,and a Venn diagram was drawn and designed.Cytoscape software was used to plot and design the final intersection results into a"active components-targets"network distribution map.A protein-protein interactions(PPI)network distribution map was created using the STRING database.The gene ontology(GO)and Kyoto encyclopedia of genes and genomes(KEGG)research analysis were conducted using the database for annotation,visualization and integrated discovery(DAVID).Results The network pharmacology results of this study showed that the critical chemical component of Jianpi Wenyang prescription in treating PPE is quercetin,and the target genes are Jun,MAPK3,MAPK1,ESR1,FOS,etc.The critical pathways are the PI3K-Akt signaling pathway and the cAMP signaling pathway.Conclusion The use of network pharmacology methods to explore the main mechanism of action and common action targets of Jianpi Wenyang prescription in the treatment of PPE provides a theoretical reference for revealing its chemically active effective components.
