Study on anti-inflammatory effect of total flavonoids from Podocarpus macrophyllus seeds
Objective To study the anti-inflammatory effect of total flavonoids from Podocarpus macrophyllus seeds.Methods Three acute inflammatory experimental models were established,namely,carrageenin-induced mouse foot swelling,xylene-induced mouse ear swelling,and lipopolysaccharide(LPS)-induced mouse inflammation.Each model was randomly divided into a blank control group,a aspirin group(200 mg/kg of aspirin),a low-dose group(60 mg/kg of total flavonoids from the seeds of Podocarpus macrophyllus),and a high-dose group(180 mg/kg of total flavonoids from the seeds of Podocarpus macrophyllus).The last two were added to a model group with 10 mice in each group.The drug was administered by gavage once a day for 7 consecutive days.In the experimental model of carrageenan-induced mouse foot swelling,the right hind foot of the mouse was injected with carrageenan to cause inflammation,while the left foot was not injected.The left and right feet of the mouse were weighed and the degree of foot swelling and swelling inhibition rate were calculated.In the xylene-induced mouse ear swelling experimental model,the right ear of the mouse was inflamed by applying xylene,while the left ear was not applied.The left and right ear slices of the mouse were weighed,and the degree of ear swelling and swelling inhibition rate were calculated;took blood again and used enzyme-linked immunosorbent assay(ELISA)to detect the levels of interleukin-1β(IL-1β)and interleukin-8(IL-8)in serum.In the LPS-induced mouse inflammation model,except for the blank control group,all other groups of mice were intraperitoneally injected with LPS to induce inflammation.Blood was collected and levels of tumor necrosis factors in serum such as tumor necrosis factor α(TNF-α),interleukin-6(IL-6)and interleukin-10(IL-10)levels were detected by ELISA.Results The inhibition rates of the low and high-dose groups of total flavonoids from Podocarpus macrophyllus seeds on the foot swelling of mice induced by carrageenin were 12.67%and 14.49%,respectively,and the inhibition rates on the ear swelling of mice induced by xylene were 15.88%and 47.65%,respectively.It had little effect on IL-1β and IL-8(P>0.05),but could significantly reduce TNF-α and IL-6 levels,and increase IL-10 level(P<0.05).Conclusion The total flavonoids of Podocarpus macrophyllus seeds can inhibit the swelling of the feet and ears of mice and have certain anti-inflammatory effects,which may be related to the reduction of serum TNF-α and IL-6 levels,and increase in IL-10 level.
