Exploring the mechanism of Yuhuo cough formula in treating cough based on network pharmacology
Objective To construct a network relationship between the pharmacological components,cough,targets,and pathways of Yuhuo cough formula using network pharmacology,and to study the mechanism of action of Yuhuo cough formula in relieving cough.Methods The traditional Chinese medicine systems pharmacology database and analysis platform(TCMSP)was used to search for the pharmacological components and corresponding targets of the Yuhuo cough formula.Cough-related targets were obtained from Drugbank,GeneCards,PharmaGkb,and Therapeutic Target Database(TTD),and online software genedenovo was used to screen the intersection targets of the drug and disease effects.Based on the DAVID database,gene ontology(GO)function and Kyoto encyclopedia of genes and genomes(KEGG)pathway enrichment analysis were conducted on relevant gene targets.Protein-protein interaction networks were established using the STRING 11.0 online database.The active ingredient-disease target relationship networks were plotted using Cytoscape.Results There are 205 active ingredients in the Yuhuo cough formula,mainly flavonoids.The key targets for treating cough include cytochrome P4503A4(CYP3A4),histone deacetylase(HDAC),estrogen receptor 1(ESR1),etc.After GO enrichment analysis,it is found that there are organic matter metabolism processes and chemical reactions related to biological processes(BP),organelles,cytoplasm and membrane boundary organelles related to cellular components(CC),and ion binding,organic cyclic compounds and heterocyclic compounds binding related to molecular functions(MF).The Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway mainly involves metabolic pathways,pathways in cancer,steroid hormone biosynthesis,and serotonin synapse.Conclusion The main components of the Yuhuo cough formula are flavonoids,achieving anti-inflammatory and cough-relieving effects mainly by inhibiting the expression of inflammatory factors.
