口腔黏膜下纤维化大鼠模型造模方法的优化

目的 比较三种口腔黏膜下纤维化(OSF)大鼠模型造模方法:槟榔碱涂擦、颊黏膜下注射槟榔碱、槟榔碱涂擦联合颊黏膜下注射槟榔碱,探讨建立大鼠OSF模型的更优造模方法。方法 选择雄性SPF级SD大鼠 40 只,随机分成空白对照组、槟榔碱涂擦组、槟榔碱注射组、槟榔碱涂擦+注射组各 10 只。观察处理后每周各组大鼠的生存状况、张口度及体重变化;处理 8 周后处死大鼠,取颊黏膜进行苏木精-伊红(HE)染色观察颊黏膜组织炎症变化、免疫组化检验颊黏膜组织中转化生长因子-β1(TGF-β1)阳性程度及蛋白质印迹法(WB)检测颊黏膜组织中TGF-β1 蛋白表达量。结果 各组大鼠经过 8 周造模后,双颊部颊黏膜出现不同程度发白、呈纤维条索样改变;其中,槟榔碱涂擦组大鼠双颊黏膜下纤维化程度不一、分布不均。槟榔碱涂擦组大鼠体重与空白对照组比较,差异无统计学意义(P>0。05),槟榔碱注射组、槟榔碱涂擦+注射组大鼠体重均下降(P<0。05),其中槟榔碱涂擦+注射组大鼠体重下降最明显(P<0。05)。与空白对照组比较,三种造模方式的OSF大鼠张口度均降低(P<0。05),与槟榔碱涂擦组比较,槟榔碱注射组和槟榔碱涂擦+注射组张口度降低更明显(P<0。05);与空白对照组比较,三种造模方式的OSF大鼠TGF-β1 表达量均显著上升(P<0。05),且各模型组间比较,差异有统计学意义(P<0。05)。其中TGF-β1 在槟榔碱涂擦+注射组中表达量最高。结论 槟榔碱涂擦联合颊黏膜下注射槟榔碱是建立大鼠口腔黏膜下纤维化的优选造模方法。
Optimization of modeling method of rat model of oral submucous fibrosis
Objective To compare three methods of establishing rat model of oral submucous fibrosis(OSF):arecoline rubbing,buccal submucosal injection of arecoline,arecoline rubbing combined with buccal submucosal injection of arecoline,so as to explore the better method of establishing rat OSF model.Methods Forty male SPF grade SD rats were randomly divided into the blank control group,the arecoline rubbing group,the arecoline injection group and the arecoline rubbing plus injection group,with 10 rats in each group.The living conditions,mouth opening and weight changes of rats in each group were observed every week after treatment.After 8 weeks of treatment,rats were sacrificed.The buccal mucosa was taken and stained with hematoxylin-eosin(HE)to observe the inflammatory changes of buccal mucosa,the positive degree of transforming growth factor-beta1(TGF-β1)in buccal mucosa was detected by immunohistochemistry,and the expression of TGF-β1 protein in buccal mucosa was detected by western blot.Results After 8 weeks of modeling,the buccal mucosa of each group showed varying degrees of whitening and fiber-like changes.Thereinto,the degree of submucosal fibrosis in both cheeks of rats in the arecoline rubbing group was different and uneven.There was no significant difference in body weight between the arecoline rubbing group and blank control group(P>0.05),but the weight of rats in the arecoline injection group and the arecoline rubbing plus injection group all decreased(P<0.05),and the weight of rats in arecoline rubbing plus injection group decreased most obviously(P<0.05).Compared with the blank control group,the mouth opening of OSF rats in the three models decreased(P<0.05),and the mouth opening of the arecoline injection group and the arecoline rubbing plus injection group decreased more obviously(P<0.05).Compared with the blank control group,the expression levels of TGF-β1 in OSF rats with three models were significantly increased(P<0.05),the differences among the model groups were also statistically significant(P<0.05),and the expression level of TGF-β1 was the highest in the arecoline rubbing plus injection group.Conclusion Arecoline rubbing combined with buccal submucosal injection of arecoline is the best method to establish oral submucous fibrosis model in rats.
