Establishment of determination method of total flavonoids in Equisetum arvense L.and optimization of extraction process
Objective To establish a method for the determination of total flavonoids in Equisetum arvense L.and optimize its extraction process.Methods The effects of extraction temperature,ultrasound power,ultrasound time,and material-liquid ratio on the total flavonoids in Equisetum arvense L.were investigated by single factor experiments.Box-Benhnken experimental design principle was used to optimize the extraction process of total flavonoids by 4 factors and 3 levels experimental design and response surface analysis.The content of total flavonoids was determined by NaNO2-Al(NO3)3-NaOH colorimetry.Results The order of influence of four factors on the content of total flavonoids in Equisetum arvense L.was material-liquid ratio>ultrasound power>ultrasound time>extraction temperature.The optimum extraction process of total flavonoids was as follows:ultrasound time as 55 min,material-liquid ratio as 1∶40 g/ml,extraction temperature as 40℃and ultrasound power as 60%(maximum power as 1000 W).Under these conditions,the contents of total flavonoids in the vegetative stem and reproductive stem of Equisetum arvense L.were 13.32 and 6.92 mg/g,respectively.Conclusion The determination method and extraction process of total flavonoids established in this experiment are simple and easy to operate,with high extraction efficiency and good stability,and are suitable for the extraction and content determination of total flavonoids in Equisetum arvense L.
