Bioinformatics analysis and preliminary verification of IncRNA LBX2-AS1 in colon cancer
Objective To explore the effect of long noncoding RNA(LncRNA)LBX2 antisense RNA 1(LBX2-AS1)on the prognosis of colon cancer and conduct preliminary verification based on bioinformatics.Methods The gene LBX2-AS1,which was closely related to the prognosis of colon cancer,was searched and identified in gene expression profiling interactive analysis(GEPIA)database.The data set of colon cancer and survival information were downloaded from UCSCxena database(TCGA-COAD)and divided into high and low expression groups of LBX2-AS1,and then the survival curve was drawn.The expression of LBX2-AS1 in clinical specimens of colon cancer and adjacent tissues was detected respectively,and its correlation with clinicopathological features of colon cancer was analyzed.After sliencing the expression of LBX2-AS1 in SW480 cell line,the proliferation,invasion,and metastasis abilities of each group of cells were detected by the cell proliferation assay,cell scratching assay and Transwell invasion assay.Results GEPIA database search results showed that LBX2-AS1 was closely related to the prognosis of colon cancer patient.The analysis of survival information of TCGA-COAD downloaded from UCSCxena database showed that with the extension of survival time,the number of survivors in the group with high expression of LBX2-AS1 was significantly less than that in the group with low expression of LBX2-AS1.Compared with the adjacent tissues,LBX2-AS1 was significantly higher in the colon cancer tissue,and the expression level was not related to the clinical parameters such as patient's age and sex,but was positively correlated with the pathological stage and lymph node metastasis of colon cancer.After LBX2-AS1 was silenced,the proliferation rate of colon cancer cell SW480 decreased significantly,the scratch distance widened significantly,and the number of invasive cells decreased significantly,with statistically significant differences(P＜0.05).Conclusion Based on bioinformatics analysis,in combination with clinical data and cell experiments,it can be concluded that LncRNA LBX2-AS1 plays a role in promoting cancer in colon cancer.
