目的 研究精液中的巨噬细胞及抗精子抗体阳性对精子质量的影响,探索小豆蔻明能否通过作用于精液中的巨噬细胞及抗精子抗体而发挥保护精子的作用。方法 用Percoll梯度离心法得优质精子,将其与不同浓度的巨噬细胞共培养,筛选出对精子有损伤的巨噬细胞浓度范围;再将含有该损伤浓度范围的巨噬细胞、抗精抗体阳性的精子悬液,分别单独及两者均有与一定量的小豆蔻明共培养,观察结果。结果 加有巨噬细胞且浓度达 0。6×106/ml及以上的精子悬液组(高浓度组)精子的存活时间明显比空白组(精子悬液组)短;含有高浓度巨噬细胞的抗精子抗体阳性精子悬液组与空白组比较,精子的存活时间显著降低,差异有统计学意义(P<0。01)。小豆蔻明对巨噬细胞、抗精子抗体及两者结合对精子的损伤有保护作用,可提升精子存活率(P<0。01)、减少精子DNA碎片率及畸形率(P<0。05)。结论 一定浓度巨噬细胞、抗精子抗体及两者共存可以明显降低精子存活数目;初步研究表明,小豆蔻明可逆转巨噬细胞及抗精子抗体对精子的损伤作用。
Research on the impact of cardamonin on sperm macrophages and anti-sperm antibodies
Objective To study the impact of sperm macrophages and anti-sperm antibodies positive on sperm quality,and to investigate whether cardamonin can protect sperm by acting on sperm macrophages and anti-sperm antibodies.Methods High-quality sperm was obtained by Percoll gradient centrifugation,and it was co-cultured with macrophages of different concentrations to screen out the concentration range of macrophages that were harmful to sperm.Then,macrophages and sperm suspensions with positive anti-sperm antibodies containing the damage concentration range were co-cultured with a certain amount of cardamonin separately and their combination,and the results were observed.Results The survival time of sperm in the sperm suspension group(the high concentration group)with macrophages and the concentration of 0.6×106/ml or above was significantly shorter than that in the blank group(the sperm suspension group).Compared with the blank group,the survival time of sperm in the anti-sperm antibodies positive sperm suspension group containing high concentration macrophages was significantly reduced,with statistically significant difference(P<0.01).In addition,cardamonin had a protective effect on macrophages,anti-sperm antibodies and their combination in sperm damage,with statistically significant differences on improving sperm survival rate(P<0.01),and with statistically significant differences in reducing sperm DNA fragmentation rate and deformity rate(P<0.05).Conclusion A certain concentration of macrophages,anti-sperm antibodies and their coexistence can significantly reduce the number of sperm survival.The preliminary study shows that cardamonin can reverse the damage of macrophages and anti-sperm antibodies to sperm.
