Exploring the preventive protective mechanism of combined use of Xiaoyao San and green tea polyphenols on acute liver injury based on network pharmacology
Objective To explore the possible mechanism of the preventive protective effect of combined use of Xiaoyao San and green tea polyphenols on acute liver injury based on network pharmacology.Methods The chemical constituents and potential target intersections of Xiaoyao San and green tea polyphenols were screened based on network pharmacology.GeneCards and NCBI database were used to query target associated with acute liver injury.Protein-protein interactions(PPI)analysis was performed on the STRING platform,and the Kyoto encyclopedia of genes and genomes(KEGG)pathway enrichment analysis and gene ontology(GO)analysis were performed on potential targets using the DAVID database to predict the mechanism and pathway of drug combination effects.Results Xiaoyao San can act on the target of hypoxia-inducible factor(HIF)-1A through its potential key components,such as Isolicoflavonol and Isotrifoliol,start HIF-1 signaling pathway,regulate the occurrence of inflammatory reaction and play an anti-oxidative stress role.When combined with green tea polyphenols,it can enhance the effect on HIF-1A target,thus synergistically enhancing its preventive and protective efficacy.Conclusion This study preliminarily clarified the mechanism of Xiaoyao San in preventing and protecting acute liver injury through multiple components,targets,and pathways,and explored its potential synergistic enhancement effect and mechanism when used in combination with green tea polyphenols,providing a theoretical basis for optimizing the dosage scheme and developing new drugs for clinical combination of traditional Chinese medicine.
