Research on screening of anti-inflammatory active fractions of Picris darurica Fisch.and its function mechanism
Objective To screen the anti-inflammatory active fractions of Picris davurica Fisch.,so as to preliminarily investigate its function mechanism.Methods Extracts of ethanol extract of Picris davurica Fisch.with different polarity fractions of petroleum ether,ethyl acetate,n-butanol and water were prepared.RAW264.7 cells were induced by lipopolysaccharide(LPS)to establish an inflammatory model in vitro.The impacts of different polarity fractions on cell proliferation activity were detected by methyl thiazolyl tetrazolium(MTT)method,and the safe administration range was determined.Griess method and enzyme-linked immunosorbent assay(ELISA)were used to detect the contents of nitric oxide(NO),tumor necrosis factor-α(TNF-α),interleukin-6(IL-6)and interleukin-1β(IL-1β)in cell supernatant,respectively.Results There was no obvious toxicity to cells when the concentration of liquid medicine in each polarity extracts fraction of the ethanol extract of Picris davurica Fisch.was 100,50 and 25 μg/ml.Compared with the model group,the ethyl acetate fraction(the high,medium and low dose groups)of the ethanol extract of Picris davurica Fisch.could all significantly reduce the secretion of inflammatory factors NO,TNF-α,IL-6 and IL-1β(P<0.01),and showed a dose-dependent relationship.The secretion of NO,TNF-α,IL-6 and IL-1β was not significantly inhibited in the petroleum ether fraction group,the n-butanol fraction group and the water fraction group.Conclusion The active fraction of anti-inflammatory effect of ethanol extract of Picris davurica Fisch.is mainly concentrated in ethyl acetate fraction,and its function mechanism is by inhibiting the secretion of NO,TNF-α,IL-6 and IL-1β.
