Effect of ivabradine hydrochloride on ventricular remodeling in rats with adriamycin-induced heart failure
Objective To investigate the effect of ivabradine on ventricular remodeling in rats with adriamycin-induced heart failure.Methods 50 specific pathogen-free grade 9-week-old Wistar rats(weighing about[300±10]g,male and female)were divided into control group(10 rats),model group(20 rats),and treatment group(20 rats)using a randomized numerical table method.Adriamycin-induced method was used to construct a rat heart failure model,and left ventricular ejection fraction(LVEF)35%-50%was taken as the criterion for model completion.Ivabradine hydrochloride(10 mg/[kg·d])was given in the treatment group,and an equal amount of saline was given in the control and model group.After 4 weeks,changes in left ventricular end-systolic diameter(LVEDs),left ventricular end-diastolic diameter(LVEDd),LVEF,and left ventricular fractional shortening(LVFS)were recorded by cardiac ultrasound.Hematoxylin-eosin staining was performed to observe the left ventricular cardiomyocytes,and Luminex,protein immunoblotting,and enzyme-linked immunosorbent assay were used to detect the levels of monocyte chemotactic protein 1(MCP-1),tumor necrosis factor α(TNF-α),brain natriuretic peptide(BNP),interleukin 1β(IL-1β),granulocyte colony-stimulating factor(G-CSF),and other factors.Results Compared with the control group,LVEDs and LVEDd were increased(P<0.05)and LVEF and LVFS were decreased in the model group(P<0.05),and the expression levels of factors such as MCP-1,TNF-α,IL-1β,G-CSF and BNP were increased(P<0.05).Compared with the model group,LVEDs and LVEDd were reduced(P<0.05)and LVEF and LVFS were increased in the treatment group(P<0.05).and the levels of the above factors were reduced(P<0.05).The morphology of cardiomyocytes in the control group was normal,and cardiomyocytes in the model group were obviously abnormal with inflammatory cell infiltration,which was partially relieved after treatment.Conclusion The method of intraperitoneal injection of amiodarone has a high and stable success rate in preparing a rat heart failure model.Ivabradine hydrochloride can effectively inhibit the inflammatory response in this model and improve ventricular remodeling.
