Intervention mechanism of flavonoids from Brassica juncea on colonic mucosa of UC rats
Objective To explore the intervention mechanism of flavonoids from Brassica juncea on colonic mucosa in ulcerative colitis(UC)rats.Methods A total of 45 SD rats were divided into a blank group(10 rats)and a modeling group(35 rats).The modeling group was randomly divided into a model group,a low and a high concentration group of flavonoids in Brassica juncea according to random number table.After treatment,the disease activity index(DAI)score,colonmucosa damage index(CMDI)score,colonic tissue pathology examination were performed;and the contents of tumor necrosis factor(TNF)-α,interleukin(IL)-1β and IL-10 in colonic mucosa of rats were detected by enzyme-linked immunosorbent assay,and the expressions of mucin 2(MUC2)and epidermal growth factor(EGFR)were detected by immunohistochemistry.Results Compared with model group,the DAI score,CDMI score,and the contents of TNF-αand IL-1β of the low and the high concentration group of flavonoids in Brassica juncea decreased(P<0.05),while the content of IL-10,the expression levels of MUC2 and EGFR increased(P<0.05).Compared with the low concentration group,the DAI score,CDMI score,and the contents of TNF-α and IL-1β decreased in high concentration group(P<0.05),while the content of IL-10,the expression levels of MUC2 and EGFR increased in high concentration group(P<0.05).HE staining results showed that the degree of colonic tissue lesions of rats in both low and high concentration groups were improved and the pathological damage results were reduced,especially in high concentration group.Conclusion Flavonoids from Brassica juncea can regulate the intestinal epithelial system by regulating the levels of TNF-α,IL-1β and IL-10 in the colonic mucosa of UC rats and increasing the expression of MUC2 and EGFR proteins,so as to achieve the purpose of repairing the colonic mucosa.
