Study on the pharmacological mechanism of Zhengchang San in intervening functional gastric emptying disorders in rabbits based on the micro dynamic structure of the stomach
Objective To explore the possible pharmacological mechanism of acupoint application of Zhengchang San in improving intervention for functional delayed gastric emptying(FDGE).Methods A total of 30 male rabbits were randomly divided into a normal group,a model group,and an acupoint application group,with 10 rabbits in each group.The normal group underwent sham surgery,while the rabbits in model group and acupoint application group underwent subtotal gastrectomy for modeling.The normal group and the model group were given non-medicine acupoint application,while the acupoint application group was treated with acupoint application of Zhengchang San.Each group was intervened for 14 days.The levels of potassium(K+)and motilin(MTL)in blood of rabbits were detected before and on the 15th day after surgery.On the 16th day after surgery,the amount of residual pigment in the stomach was measured to reflect residual gastric emptying function;on the 16th day after surgery,the ultrastructure of residual gastric smooth muscle cells and interstitial cells of cajal(ICC)was determined using electron microscopy.Results Compared with the normal group,the levels of MTL and K+in the model group were significantly reduced(P<0.001).Compared with the model group,the levels of MTL and K+in the acupoint application group were significantly increased(P<0.001).Compared with the normal group,the amount of residual pigments in the stomach of model group increased(P<0.001).Compared with the model group,the amount of residual pigments in the stomach of acupoint application group was significantly reduced(P<0.001).Compared with the normal group,the model group had disordered nuclear arrangement,irregular morphology,blurred ICC cell structure,nuclear condensation,fewer mitochondria,and the appearance of vacuoles.Compared with the model group,the acupoint application group had less cell damage,more orderly arrangement,clearer ICC cell structure,increased protrusions,and no autophagosomes,indicating that acupoint application of Zhengchang San can improve the damage of rabbit gastric mucosal cells and improve the ultrastructure of ICC in smooth muscle cells of rabbits after subtotal gastrectomy.Conclusion Acupoint application of Zhengchang San can effectively prevent the occurrence of FDGE after subtotal gastrectomy in rabbits.Its mechanism of action may be related to stabilizing K+levels,promoting MTL secretion,improving gastric emptying function,and improving the ultrastructure of residual gastric smooth muscle cells and ICC.
