目的 探讨采用两种超排卵剂量下的鼠胚实验对辅助生殖实验室进行质控的可行性.方法 对SPF级昆明雌性小白鼠行腹腔注射孕马血清促性腺激素(PMSG)和绒毛膜促性腺激素(HCG)超排卵,在采用8 IU PMSG+8 IU HCG和16IU PMSG+16IU HCG两种不同注射剂量下进行体外受精和体内受精,观察不同剂量、不同受精方式下鼠胚发育的优胚率和囊胚形成率.结果 分别实施8 IU PMSG+8 IU HCG超排卵剂量与16 IU PMSG+16 IU HCG超排卵剂量后,雌性小鼠体外受精,鼠胚优胚率分别为90.965%和81.877%,囊胚形成率分别为82.341%和75.081%,差异有统计学意义(P<0.05);雌性小鼠体内受精,鼠胚优胚率分别为93.096%和85.889%,囊胚形成率分别为83.432%和76.994%,差异有统计学意义(P<0.05).结论 采用两种超排卵剂量下的鼠胚实验对辅助生殖实验室进行质控具有可行性,恰当的超排卵剂量,能获得较高的优胚率和囊胚形成率.
Investigation on quality control of assisted reproductive laboratory by mouse embryo experiment with two doses of superovulation
Objective To investigate the feasibility of quality control of assisted reproductive laboratory by mouse embryo experiment with two doses of superovulation.Methods Female mice of SPF grade in Kunming were injected intraperitoneally with pregnant mare serum gonadotropin(PMSG)and human chorionic gonadotropin(HCG)for superovulation.In vitro fertilization and in vivo fertilization were carried out at different injection doses of 8 IU PMSG+8 IU HCG and 16 IU PMSG+16 IU HCG.The excellent embryo rate and blastocyst formation rate of mouse embryo development under different doses and different fertilization methods were observed.Results After 8 IU PMSG+8 IU HCG superovulation dose and 16 IU PMSG+16 IU HCG superovulation dose were applied respectively,female mice were fertilized in vitro,and the excellent embryo rate of mouse embryos were 90.965%and 81.877%,and the blastocyst formation rates were 82.341%and 75.081%,respectively,with statistically significant differences(P<0.05).After in vivo fertilization of female mice,the excellent embryo rates of mouse embryos were 93.096%and 85.889%,and the blastocyst formation rates were 83.432%and 76.994%,respectively,with statistically significant differences(P<0.05).Conclusion It is feasible to control the quality of assisted reproductive laboratory by using mouse embryo experiments at two doses of superovulation.Appropriate doses of superovulation can obtain higher excellent embryo rate and blastocyst formation rate.
