氨基酰tRNA合成酶在肿瘤研究中的进展

氨基酰tRNA合成酶(aaRs)的经典功能是催化氨基酸与对应的tRNA结合,形成氨基酰tRNA,参与蛋白质合成,但aaRs在进化过程中还发展出许多与氨基酰化无关的非典型功能.aaRs表达失调与肿瘤、神经系统疾病、心血管疾病等多种疾病相关.目前已有多项研究证实aaRs与结直肠癌、肺癌、乳腺癌等多种肿瘤的发生发展相关.随着aaRs与肿瘤之间的关系变得更明晰,aaRs抑制剂有希望发展成为一种新型肿瘤治疗药物.本文综述了aaRs的功能以及aaRs与肿瘤的相关性,并讨论了抗肿瘤活性的aaRs抑制剂的开发思路.
Progress of aminoacyl-tRNA synthetase in tumor research
The classic function of aminoacyl-tRNA synthetase(aaRs)is to catalyze the binding of amino acids with their corresponding tRNA,forming aminoacyl-tRNA and participating in protein synthesis.However,aaRs have also developed many atypical functions unrelated to aminoacylation during their evolution.The dysregulation of aaRs expression is associated with various diseases such as tumor,nervous system diseases,and cardiovascular diseases.At present,many studies have confirmed that aaRs are related to the occurrence and development of colorectal cancer,lung cancer,breast cancer and other tumors.As the connection between aaRs and tumors becomes clearer,aaRs inhibitors have the potential to develop into a new type of tumor therapeutic drug.In this paper,the function of aaRs and the correlation between aaRs and tumor are reviewed,and the development ideas of aaRs inhibitors with anti-tumor activity are discussed.
