SOD was extracted fromPleurotus ostreatus mycelium with direct method, toluene method and ethanol-chloroform method respectively, and the enzyme activity was tested. The results showed that all the three extraction methods can obtain SOD, and the extraction effect of ethanol – chloroform method was the best through the comparison. Extraction conditions was adding 1 g mushroom mycelium into 8ml NaHCO3 solution of 0.2 mol/L and 6ml ethanol - chloroform solution, then centrifuging 30min at 4 000 r/min after stirring 2 h. The crude enzyme activity of 1.34515 U/μg was the highest. After purifying the specific activity was 1.521 U/ mg. The purification multiple was 1.720, and the recovery rate was 91.7%.
