响应面法优化沙棘果油提取工艺的研究

以沙棘果浆为原料,采用复合酶酶解技术对沙棘果油进行提取,考察提取过程中加酶量、果胶酶与纤维素酶质量比、酶解温度、酶解时间对沙棘果油得率的影响.在单因素实验的基础上,采用Box-Behnken响应面分析法进行优化.确定最佳提取工艺条件为加酶量0.30 g(以100 g沙棘果浆计)、果胶酶与纤维素酶质量比1.54∶1、酶解温度55℃、酶解时间4.24h,在此条件下,沙棘果油得率为5.28%,比优化前提高了19.7%.
Optimization of extraction of seabuckthorn fruit oil by response surface methodology
With seabuckthorn pulp as raw material,seabuckthorn fruit oil was extracted by compound enzymatic hydrolysis technology.The effects of enzyme dosage,mass ratio of pectinase to cellulose,hydrolysis temperature and hydrolysis time on the yield of seabuckthorn fruit oil were studied.Based on single factor experiment,the extraction conditions were optimized by Box-Behnken response surface methodology as follows:enzyme dosage 0.30 g(based on 100 g seabuckthorn pulp),mass ratio of pectinase to cellulase 1.54∶1,hydrolysis temperature 55 ℃ and hydrolysis time 4.24 h.Under these conditions,the yield of seabuckthorn fruit oil was 5.28%,increasing by 19.7% than that before optimization.
