单细胞测序技术在肺癌免疫治疗中的应用

摘要
肺癌的治疗模式随着免疫检查点抑制剂的问世发生了巨大的变化,已经使许多患者生存获益.但肺癌广泛的异质性以及免疫微环境的复杂性限制了免疫治疗的应用,表现为治疗适用人群的局限、有效率和耐药的局限以及广泛且异质的免疫相关不良事件.近年来,单细胞测序技术在灵敏度、准确性和效率方面均得到提高,因其能够揭示肺癌免疫微环境的异质性、精细化探索肺癌微环境、实现患者精细分层并指导临床实践,单细胞测序技术已对肺癌的免疫治疗产生了深远影响.文章综述了单细胞测序的种类、RNA测序技术在肺癌微环境探索中的研究进展以及在肺癌免疫治疗中的应用,以期为了解肺癌的生物学行为提供新视角,为精准医学及免疫药物开发提供线索,为肺癌的个性化临床诊疗提供理论依据.
Abstract
The treatment mode of lung cancer has changed greatly with the advent of immune checkpoint inhibitors(ICIs),bringing survival benefits to many patients.However,the widespread heterogeneity of lung cancer and the complexity of the immune microenvironment limit the application of immunotherapy,which is manifested in the restricted eligibility of patients,response rate,and the heterogeneous range ofimmune-related adverse events.The sensitivity,accuracy and efficiency of single-cell sequencing technology have been improved in the past decade.This technology is now capable of revealing the heterogeneity of the lung cancer immune microenvironment,allowing for a more refined exploration of the lung cancer microenvironment,enabling precise patient stratification,and guiding clinical practice.This technology has had a profound impact on the immunotherapy of lung cancer.This article reviews the types of single cell sequencing,the progress of RNA sequencing in the exploration of lung cancer microenvironment and the application of lung cancer immunotherapy.This paper provides new perspectives for understanding the biological behavior of lung cancer,clues for precision medicine and immunotherapy development,and a basis for personalized clinical diagnosis and treatment of lung cancer.
