新银灵颗粒对寻常型银屑病血热证患者外周血Th22细胞及其细胞因子的影响

摘要
目的 观察新银灵颗粒对寻常型银屑病(PsV)血热证患者外周血辅助性T细胞 22(Th22)及其细胞因子的影响.方法 选取 2019 年 12 月—2020 年 12 月在天津市中医药研究院附属医院皮肤科门诊就诊的 30 例PsV血热证患者为研究组,10 例健康志愿者为对照组.研究组采用新银灵颗粒治疗8周,分别于治疗前后采取外周血,流式细胞术检测外周血Th22 细胞比例,酶联免疫吸附测定(ELISA)法检测白细胞介素-22(IL-22)水平,实时荧光定量聚合酶链反应(RT-qPCR)检测IL-22 mRNA表达.结果 经过新银灵颗粒治疗,研究组与治疗前比较,银屑病面积及严重程度指数(PASI)评分、中医证候评分和皮肤病生活质量指数评分(DLQI)均显著下降(均P<0.01);治疗后外周血中Th22 细胞比例、IL-22 及IL-22 信使RNA(mRNA)表达水平均显著降低(P<0.01).结论 新银灵颗粒可能通过抑制Th22 细胞活化、降低Th22 细胞因子表达,从而治疗PsV血热证.
Abstract
Objective In order to observe the effects of Xinyinling granule on T helper cell 22(Th22)and its cytokines in peripheral blood of patients with blood heat syndrome of psoriasis vulgaris(PsV).Methods Thirty patients with blood heat syndrome of PsV treated in the dermatology department of Tianjin academy of traditional Chinese medicine affiliated hospital from December 2019 to December 2020 were selected as the study group,and 10 healthy volunteers were taken as the control group.The study group was treated with Xinyinling granules for eight weeks.Before and after treatment,the proportion of Th22 cells in peripheral blood was detected by flow cytometry,interleukin-22(IL-22)level was detected by enzyme-linked immunosorbent assay(ELISA),and IL-22 mRNA expression level was determined by real-time fluorescence quantitative polymerse chain reaction(RT-qPCR).Results After treating with Xinyinling granule,psoriasis area and severity index(PASI),Chinese medicine symptom complex score,and dermatology life quality index(DLQI)in the study group were significantly lower than before treatment(all P<0.05).The proportion of Th22 cells,the expression levels of IL-22 and IL-22 mRNA in peripheral blood of the study group were significantly decreased after treatment(P<0.01).Conclusion Xinyinling granules may treat PsV by inhibiting the activation of Th22 cells and down-regulating the expression of its cytokines.
