Objective To investigate the effects of tannic acid from gallnut extract on proliferation,migration and invasion of EJ cells and T24 cells of bladder cancer and its mechanism.Methods CCK-8 and cell counting assay were used to detect the inhibitory effect of tannic acid at different concentrations on the proliferation of EJ and T24 cells.Plate cloning,cell scratch and Transwell assay were used to detect the effects of tannic acid on the proliferation,migration and invasion of EJ and T24 cells.The effects of tannic acid on apoptosis,metastasis and expression of retinoic acid-associated orphan nuclear receptor C protein(RORC)in bladder cancer EJ and T24 cells were detected by Western Blot assay.Results Tannic acid at different concentrations inhibited the proliferation of EJ and T24 cells in a dose-dependent manner(P<0.05),and the proliferation,migration and invasion of EJ and T24 cells decreased with the increase of tannic acid concentration(P<0.05).Tannic acid up-regulated the expression of apoptosis-related proteins PARP1,Caspase7 and E-cadherin in bladder cancer EJ and T24 cells.Meanwhile,the expression of transfer-related proteins N-cadherin,MMP-2,Vimentin and SNAI1 was down-regulated.The expression of EJ and T24 apoptosis-related proteins correlated with the up-regulation of RORC.Conclusion Tannic acid inhibited the proliferation and migration of bladder cancer cells EJ and T24 by up-regulating RORC expression.
