Protective Effect of Puerarin on the Heart of D-galactose Induced Aging Mice
Objective To investigate the protective effects of puerarin on D-galactose(D-gal)induced heart aging.Methods 24 Kunming mice aged 6-8 week old were randomly divided into normal control group,D-gal treated group,low-dose puerarin-treated group and high-dose puerarin-treated group.The aging model was established by continuous subcutaneous injection of D-gal(150 mg/kg/d),puerarin and the same amount of 0.9%sodium chloride solution were given intragastric administration after successful modeling.After 60 days,HE staining was performed to observe morphological changes of myocardial tissues,automatic biochemical analyzer was used to detect serum biochemical parameters including the cardiac troponin T(cTnT)and creatine kinase(CK),malondialdehyde(MDA)content and superoxide dismutase(SOD)activity were measured by ELISA,fluorescence probe dihydroethidium(DHE)were used to detect the levels of reactive oxygen species(ROS),immunohistochemical method was used to detect the expression of 8-hydroxydeoxyguanosine(8-OHdG),Western blot was used to detect the expressions of 4-hydroxy-2-nonenal(HNE),p21 and p53 in myocardial tissues.Results Compared with control group,irregularly arranged cardiac muscle fibers were observed in myocardial tissues of D-gal-treated mice,the levels of cTnT,CK,ROS and MDA were upregulated,and the SOD activity was significantly decreased,the expressions of 8-OHdG,4HNE,p21,and p53 was upregulated after D-gal treatment(P<0.01).Compared with D-gal treated group,serum cTnT and CK levels in low-dose puerarin-treated group and high-dose puerarin-treated group were significantly decreased(P<0.05).In addition,the expression levels of ROS,MDA,8-OHdG,4-HNE,p21 and p53 in myocardial tissues were significantly decreased,while the level of SOD was significantly increased(P<0.05).Conclusion Puerarin might serve as a potential effective Chinese herbal active constituent for anti-aging therapy.
