Comparative Study on Uterine Vascular Architecture of Kidney Deficiency Three Syndrome Rat Models
Objective To investigate the effect of three syndromes of deficiency of kidney essence,deficiency of kidney Yin and deficiency of kidney Yang on uterine angioarchitecture in female rats.Methods Female SD rats were randomly divided into four groups,control group,kidney essence deficiency group,kidney Yin deficiency group and kidney Yang deficiency group,12 rats in each group.The three kidney deficiency models were established by repeated superovalation for 5 times,intragastric administration of thyroid tablets combined with serpine suspension for 21 days,and intragastric administration of hydroxyurea suspension at 4℃for 21 days.After 15 days observation,the rats were sacrificed at the late stage of estrus.Blood and uterus were collected from 8 rats randomly selected from each group.Serum follicle-stimulating hormone(FSH),anti-meulerian hormone(AMH),estrogen(E2),progesterone(P)levels were measured by ELISA.The endometrial thickness was measured and the number of glands was counted by HE staining.The remaining rats were perfused with barium sulfate solution.Uterine angioarchitecture was observed and quantified by Micro-CT.Results Compared with the control group,the content of FSH in serum of the three model groups was significantly increased(P<0.01).The content of serum E2,P and AMH was significantly decreased,and the vascular volume proportion of the three models was decreased as well(P<0.05).In kidney essence deficiency group,the endometrial thickness,the number of endometrial gland,and the number of second,third and fourth blood grade vessels in uterus was significantly decreased(P<0.05).In kidney Yin deficiency group,abnormal uterine bleeding was observed,the mean diameter of primary and secondary grade blood vessels decreased,and the number of secondary,tertiary and quaternary grade blood vessels decreased significantly(P<0.05).In kidney Yang deficiency group,the endometrial thickness,the number of endometrial gland,and the average diameter of uterine blood vessels at all levels decreased(P<0.05).Conclusion The morphological changes and decreased receptivity of endometrium in rats were observed in three kidney deficiency models.The changes of uterine angioarchitecture may be one of the reasons.
