Effect of Electroacupuncture with"Tianshu"and"Tianshu"Combined with"Shangjuxu"on Behavior and Colon Function in Rats with Irritable Bowel Syndrome
Objective To observe and compare the effects of electroacupuncture"Tianshu"(ST25)and"Tianshu"combined with"Shangjuxu"(ST37)on the mood and colon function of rats with irritable bowel syndrome(IBS)model.To observe and compare the effects of EA-ST25,EA-ST25+ST37 on the mood and colon function of rats modeling IBS,and to preliminarily investigate whether there is any synergistic or superimposed effect of ST25 and ST37.Methods 55 male clean-grade Wistar rats,were randomly divided into Control(10 rats),model groups(45 rats).The model rats were intervened for 10 days by the combination of chronic and acute stress;After the end of modeling,male Wistar rats were randomly divided into model(8 rats),EA-ST25(8 rats),EA-ST25+ST37 groups(8 rats).The two treatment groups received continuous EA interventions for 14 days,and at the end of the intervention,each group of rats was subjected to the elevated cross-maze test,the determination of colonic pain threshold,the measurement of serum 5-hydroxytryptamine(5-HT)and the expression of 5-hydroxytryptamine 4 receptor(5-HT4 R)protein in the colon.Results Compared with the model group,the percentage of entries into the open arm and time increased and were statistically significant(P<0.01)in the EA-ST25 and the EA-ST25+ST37.The visceral pain sensitivity thresholds of the ST25 and the ST25+ST37 were significantly higher and statistically significant(P<0.01).The serum 5-HT contents of the EA-ST25 and the EA-ST25+ST37 decreased(P<0.05).The relative 5-HT4 R receptor protein expression in the EA-ST25 or EA-ST25+ST37 increased(P<0.05);There was no statistically significant difference between the two treatment groups.Conclusion EA-ST25、ST25+ST37 group can both regulate the abnormalities of emotional and psychological behavior and improve the symptoms of disordered colon function in rats,but compared with EA-ST25,EA-ST25+ST37 did not show a significant synergistic effect.
