葛根菊苣茯苓方对高尿酸血症大鼠的作用与机制研究

目的 通过建立高尿酸血症(hyperuricemia,HUA)大鼠模型,探讨葛根菊苣茯苓方的降尿酸作用与可能机制.方法 将42 只SD雄性大鼠,随机分为正常组、模型组、葛根菊苣茯苓方低、中、高剂量(6.15 g/kg、12.3 g/kg、24.6 g/kg)组,别嘌醇组(0.03 g/kg),正常+葛根菊苣茯苓方组(正常给药组).除正常组与正常给药组外,其余各组大鼠每日灌胃氧嗪酸钾 1.5 g/kg、腺嘌呤 0.1 g/kg建立HUA模型,连续21 d.从第 8 天开始,除正常组与模型组外,其余各组大鼠每日灌胃葛根菊苣茯苓方及别嘌醇干预治疗,持续 14 d.造模结束后,采集大鼠血液,测定血尿酸(serum uric acid,SUA)、血肌酐(serum creatinine,SCr)、血尿素氮(blood urea nitrogen,BUN)、腺苷脱氨酶(adenosine deaminase,ADA)、黄嘌呤氧化酶(xanthine oxidase,XOD)、谷草转氨酶(aspartate aminotransferase,AST)、谷丙转氨酶(alanine aminotransferase,ALT)含量;取大鼠肝脏及肾脏,计算肾脏系数;苏木精-伊红(hematoxylin-eosin staining,HE)染色观察肾脏、肝脏病理变化;Western blot检测肾脏中有机阴离子转运体(organic anion,OAT)1、OAT3、尿酸转运体(recombinant urate transporter,URAT)1、三磷酸腺苷结合转运蛋白(ATP-binding cassette sub-family G member,ABCG)2、葡萄糖转运蛋白(glucose transporter type,GLUT)9 蛋白表达.结果 与正常组比较,模型组大鼠肾脏表面皱缩,色泽变浅,肾小管代偿性扩张,可见尿酸盐结晶;肾脏质量增加(P<0.05),肾脏系数增大(P<0.05);血清SUA、BUN、XOD、SCr、ADA含量与肾脏组织URAT1、GLUT9 蛋白表达显著上升(P<0.05);ABCG2、OAT1、OAT3 蛋白表达显著降低(P<0.05).与模型组比较,治疗组大鼠上述指标有明显改善,且差异有统计学意义(P<0.05).结论 葛根菊苣茯苓方可以通过调节尿酸盐转运相关蛋白降低HUA大鼠的高尿酸水平.
Study on the Effect and Mechanism of Gegen Juju Fuling Formula on Hyperuricemia Rats
Objective To investigate the uric acid-lowering effect and possible mechanism of Gegen Juju Fuling formula by establishing a model of hyperuricemia(HUA).Methods A total of 42 male SD rats were randomly divided into normal group,model group,Gegen Juju Fuling formula low,medium,high dose group(6.15 g/kg,12.3 g/kg,24.6 g/kg),allopurinol group(0.03 g/kg),normal+Gegen Juju Fuling formula(normal administration group).Except for the normal group and the normal administration group,rats in other groups were intragastric with potassium oxazinate of 1.5 g/kg and adenine of 0.1 g/kg daily to establish HUA model for 21 consecutive days.Starting from the 8th day,except for the normal group and the model group,the rats in the other groups were intragastricated with Gegen Juju Fuling prescription and allopurinol every day for 14 days.The blood of the rats was collected after the modeling.Serum uric acid(SUA),blood urea nitrogen(BUN),serum creatinine(SCr),xanthine oxidase(XOD),adenosine deaminase(ADA),and alanine aminotransferase(ALT),aspartate aminotransferase(AST)were determined.The liver and bilateral kidneys of rats were taken,and the kidney coefficient was calculated.Pathological changes in the kidney and liver were observed by hematoxylin-eosin(HE)staining.Western blot was used to detect organic anion(OAT)1,OAT3,ATP-binding cassette sub-family G member(ABCG)2,recombinant urate transporter(URAT)1,glucose transporter type(GLUT)9 protein expression.Results Compared with the normal group,the kidney of the model group was larger,the surface was wrinkled and uneven,the color was lighter,the renal tubules were compensatory dilated,and urate crystals and vacuoles were visible.Body mass was significantly decreased(P<0.05),kidney mass was significantly increased(P<0.05),and kidney coefficient was significantly increased(P<0.05).The contents of serum SUA,BUN,SCr,XOD,and ADA were significantly increased(P<0.05).OAT1,OAT3,ABCG2 protein expressions decreased significantly(P<0.05),and URAT1,GLUT9 protein levels increased significantly(P<0.05).Compared with the model group,the rats in the treatment group showed significant improvement in the above indexes,and the difference was statistically significant(P<0.05).Conclusion Gegen Juju Fuling formula can improve the level of hyperuric acid in HUA rats by regulating the protein related to uric acid regulation.
