续断种子方对少精子症模型大鼠睾丸生精细胞增殖分化的影响

目的 基于丝裂原活化蛋白激酶p38/细胞外调节蛋白激酶 1/2(p38 mitogen-activated protein kinase/extracellular regulated protein kinases1/2,p38 MAPK/ERK1/2)通路探索续断种子方对少精子症模型大鼠睾丸生精细胞增殖分化的影响.方法 按随机数字表法将44只雄性SD大鼠分为4组(正常组、模型组、左卡尼汀组和续断种子方组),每组 11 只.正常组常规喂养,其余 3 组大鼠腹腔注射环磷酰胺进行造模.造模成功后,各组大鼠给予相应药物灌胃,给药 4 周后,检测大鼠精子浓度、活率及睾丸组织病理,生化法检测睾丸组织超氧化物歧化酶(superoxide dismutase,SOD)、丙二醛(malondialdehyde,MDA)、谷胱甘肽(glutathione,GSH)水平,RT-PCR与Western blot检测p38 MAPK、ERK1/2 表达.结果 与正常组比较,模型组大鼠精子浓度与精子活率明显下降(P<0.01),睾丸曲细精管分布疏松伴萎缩,各级精子细胞排列层次紊乱、数量较少,SOD、GSH水平下降(P<0.01),MDA水平升高(P<0.01),p38 MAPK、ERK1/2 表达明显升高(P<0.01);与模型组比较,续断种子方组大鼠精子浓度、活率均升高(P<0.01),睾丸曲细精管结构基本恢复正常,管壁各级生精细胞层次基本整齐,管腔内见大量成熟精子,SOD、GSH水平升高(P<0.01),MDA水平降低(P<0.01),p38 MAPK、ERK1/2 表达降低(P<0.05).结论 续断种子方能通过降低p38 MAPK、ERK1/2 表达,抑制氧化应激及调节生精细胞凋亡,促进睾丸中精子发生,从而治疗少精子症.
Effect of Xuduan Zhongzi Formula on the Proliferation and Differentiation of Testicular Germ Cells in Oligospermia Model Rats
Objective To explore the effect of Xuduan Zhongzi formula on proliferation and differentiation of testicular spermatogenic cells in oligospermia model rats based on the p38 MAPK/ERK1/2 pathway.Methods 44 male SD rats were randomly divided into 4 groups,including normal group,model group,levocarnitine group,and Xuduan Zhongzi formula group,with 11 rats in each group.The rats in normal group were fed regularly,while the rats in other three groups were induced oligospermia model by intraperitoneal injection of cyclophosphamide.After successful modeling,the rats were given corresponding therapeutic drugs by gavage.After 4 weeks of administration,the testis and epididymis tissues of rats in each group were removed to detect sperm concentration,motility and testicular histopathology.SOD activity,MDA content and GSH content in testicular tissue were detected by biochemical method.Real-time PCR and Western blot were used to detect the expressions of p38 MAPK and ERK1/2.Results Compared with the normal group,the sperm concentration and motility of rats in model group were significantly decreased(P<0.01),the distribution of seminiferous tubules in the testis was loose and atrophic,the arrangement of sperm cells at all levels was disordered and the number was less,the levels of SOD and GSH were decreased(P<0.01),while MDA was increased(P<0.01),and the expression of p38 MAPK and ERK1/2 were obviously increased(P<0.01).Compared with the model group,the sperm concentration and motility of the Xuduan Zhongzi formula group were increased(P<0.01),the structure of the testicular seminiferous tubule almostly returned to normal,the arrangement of spermatogenic cells at all levels of the tube wall was basically neat,and a large number of mature sperm were seen in the lumen.The levels of SOD and GSH increased(P<0.01),while MDA decreased(P<0.01),and the expression of p38 MAPK and ERK1/2 decreased(P<0.05).Conclusion Xuduan Zhongzi formula can inhibit oxidative stress and apoptosis of spermatogenic cells by reducing the expression of p38 MAPK and ERK1/2,and promote the development of sperm,so as to treat oligospermia.
