Experimental Study on Improvement Effects of Gualou(Trichosanthes)Extract on Myocardial Fibrosis in Rats with Ischemic Cardiomyopathy
Objective:To observe the effect of Trichosanthes Extract on myocardial fibrosis in rats with is-chemic cardiomyopathy(ICM)and explore its mechanism.Methods:36 male SD rats of clean grade were ran-domly divided into normal control group,model group and Trichosanthes Extract group with each group 12 rats.The rat model was prepared by intraperitoneal injection of 20%ethyl carbamate(5 mg/kg).Trichosanthis Ex-tract group was intragaically irrigated with Trichosanthis Extract solution 4 g/kg,and normal control group and model group were intragaically irrigated with the same dose of normal saline for 4 weeks.Myocardial fibrosis inde-xes[transforming growth factor(TGF)-1β,connective tissue growth factor(CTGF),galactoagglutinin(Gal)-3]were detected by ELISA,serum oxidative stress indexes[superoxide dismutase(SOD),malondialdehyde(MDA)]were detected by chemiluminescence method,serum inflammatory factors[interleukin(IL)-6,tumor,necrosis factor(TNF)-α,C-reactive protein(CRP)]were detected by ELISA.The pathological changes of myocardial tissue in each group were observed by light microscope.Results:The levels of serum TGF-1β,CTGF and Gal-3 in model group were higher than those in the normal group,the levels of TGF-1β,CTGF and Gal-3 in the Trichosanthes Extract group were lower than those in the model group,and the diffe-rences were statistically significant(P<0.05).The serum SOD activity in the model group was lower than that of the normal group,and the MDA content was higher than that of the normal group,the serum SOD activity of the Trichosanthes Extract group was higher than that of the model group,and the MDA content was lower than that of the model group,the differences were statistically significant(P<0.05).The levels of serum TNF-α,IL-6 and CRP in the model group were higher than those in the normal group,the levels of TNF-α,IL-6 and CRP in the Trichosanthes Extract group were lower than those in the model group,and the differences were statistically significant(P<0.05).Masson staining showed that a small number of uniform collagen fibers were found around the blood vessels of the myocardium in the normal group.A large number of collagen fibers were found in the myocardium of the model group,and they penetrated into the intertissue space,with uneven distri-bution and disorderly shape.The myocardial tissue morphology of Trichosanthis Extract group was basically nor-mal.Conclusion:Trichosanthes Extract can reduce the degree of myocardial fibrosis in ICM rats,which is rela-ted to the mechanism of antioxidation and anti-inflammatory.
