健脾宁神颗粒对抽动障碍小鼠脑组织单胺类神经递质的影响

目的:研究健脾宁神颗粒对抽动障碍小鼠脑组织单胺类神经递质的影响,以探讨健脾宁神颗粒可能的作用机制.方法:60只雄性ICR小鼠,除10只作为正常对照组外,其余各组均建立抽动障碍模型.造模成功后,将其随机分为模型对照组,健脾宁神颗粒高、中、低剂量组,硫必利阳性对照组;连续给药4周后,酶联免疫法(ELISA)检测脑组织多巴胺(DA)、5-羟色胺(5-HT)、去甲肾上腺素(NE)含量.结果:与正常对照组比较,模型对照组小鼠脑组织5-HT含量明显增加(P<0.05),DA、NE含量明显减少(P<0.05,P<0.01).与模型对照组比较,健脾宁神颗粒高剂量组小鼠脑组织5-HT含量明显减少(P<0.05),DA、NE含量明显增加(P<0.05).结论:调节脑组织单胺类神经递质DA、5-HT、NE水平是健脾宁神颗粒治疗抽动障碍的可能的作用机制之一.
Effects of Jianpi Ningshen Granule(健脾宁神颗粒)on Monoamine Neurotransmitter in Brain of Mice with Tic Disorder
Objective:To study the effect of Jianpi Ningshen Granule on the brain monoamine neurotrans-mitter in mice with tic disorder,and to explore the possible mechanism of Jianpi Ningshen Granule.Methods:60 male ICR mice,except 10 as the normal control group,other groups were established tic disorder model.Af-ter successful modeling,the rats were randomly divided into model control group,high,middle and low dose groups of Jianpi Ningshen Granule,and positive control group of Tiapride,the levels of dopamine(DA),5-hydroxytryptamine(5-HT)and noradrenaline(NE)were measured by enzyme-linked immunosorbent assay(ELISA).Results:Compared with the normal control group,the content of 5-HT in the brain of the model control group increased significantly(P<0.05),while the content of DA and NE decreased significantly(P<0.05,P<0.01).Compared with the model control group,the content of 5-HT in the brain tissue of the high-dose group was significantly decreased(P<0.05),while the contents of DA and NE were significantly in-creased(P<0.05).Conclusion:To regulate brain monoamine eurotransmitter DA,5-HT and NE levels was one of the mechanism that Jianpi Ningshen Granule on tic disorder.
