Effects of Shuizhi(Leeches)on Podocyte Injury in Focal Segmental Glomerulosclerosis Rats
Objective:To observe the effects of Leeches on podocyte injury in rats with focal segmental glo-merulosclerosis(FSGS).Methods:44 rats were randomly divided into three groups:normal group,model group,and Leech group.FSGS model was established by Doxorubicin injection in the tail vein after unilateral ne-phrectomy.Leeches group was given Leeches suspension 0.9 g/(kg·d),normal control and model groups were given equal amount of saline,they were medicated for 8 weeks.Blood triglyceride(TG)was determined,immu-nofluorescence,Western blot were used to determine Mfn2 protein expression and Mfn2 mRNA was measured by PCR after administration for 4 weeks and 8 weeks,respectively.Results:The TG of the rats in the Leeches group was lower than that in the model group(P<0.05).The expressions of Mfn2 protein and Mfn2 mRNA in the Leeches group were higher than those in the model group(P<0.05).Conclusion:Leeches can reduce the level of TG,increase the expression levels of Mfn2 protein and Mfn2 mRNA,reduce podocyte damage.
