金丝桃苷对棕榈酸诱导内皮细胞凋亡的保护作用及相关机制的实验研究

目的:研究金丝桃苷对棕榈酸诱导人脐静脉内皮细胞凋亡的影响和相关机制.方法:建立棕榈酸诱导人脐静脉内皮细胞损伤模型,实验分为正常组、棕榈酸组(50 μmol/L棕榈酸)、金丝桃苷组(50 μmol/L棕榈酸+0.1 μmol/L金丝桃苷),采用流式细胞术检测细胞凋亡情况,Western blot法检测各组细胞凋亡相关蛋白Bax(Bcl-2相关X蛋白)、Bcl-2(B淋巴细胞瘤-2蛋白)和内质网应激相关蛋白CHOP(C/EBP同源蛋白)、GRP78(葡萄糖调节蛋白78)的表达水平,PCR检测凋亡相关基因Bax、CHOP mRNA水平,划痕实验检测细胞的迁移能力.结果:与正常组比较,棕榈酸组细胞凋亡率显著增加(P<0.01),Bax/Bc-l2蛋白表达比值显著升高(P<0.01),Bax mRNA表达水平显著升高(P<0.01),迁移能力显著降低.与棕榈酸组比较,金丝桃苷组细胞凋亡率显著降低(P<0.01),Bax/Bcl-2蛋白表达比值显著降低(P<0.01),Bax mRNA表达水平显著降低(P<0.01),迁移能力显著升高(P<0.01).研究还发现,棕榈酸可提高CHOP(P<0.01)表达(P<0.01),降低GRP78表达(P<0.01);与棕榈酸组相比,金丝桃苷组CHOP和GRP78蛋白和mRNA表达水平均逆转(P<0.01).结论:金丝桃苷能够缓解棕榈酸诱导的人脐静脉内皮细胞内质网应激,减少细胞凋亡,提高细胞迁移能力.
Experimental Study on Protective Effect of Hyperoside on Palmitic Acid-Induced Endothelial Cell Apoptosis and Related Mechanism
Objective:To investigate the effect of Hyperoside on the apoptosis of Palmitic acid-induced human umbilical vein endothelial cells and the related mechanism.Methods:Human umbilical vein endothelial cells were divided into normal control group,Palmitic acid group(50 μmol/L Palmitic acid)and Hyperoside group(50pmol/LPalmitic acid+0.1 μmol/L Hyperoside).Apoptosis was detected by flow cytometry.Wes-tern blot was used to detect the levels of apoptosis related proteins(Bax and Bcl-2)and endoplasmic reticulum stress related proteins(CHOP and GRP78).The mRNA levels of Bax and CHOP were detected by PCR.Migra-tion ability of cells was detected by scratch test.Results:The apoptosis rate,Bax/Bcl-2 were significantly in-creased in the Palmitic acid group compared to those in the normal control group(P<0.01),The expression of Bax mRNA was increased in the Palmitic acid group and migration capacity was decreased compared to that in the normal control group(P<0.01).Compared with the Palmitic acid group,the apoptosis rate of the Hypero-side group was lowered(P<0.01),Bax/Bcl-2 ratio and Bax mRNA level were lower,too(P<0.01),and the migration capacity was raised(P<0.01).The results also showed that Palmitic acid could up-regulated CHOP and down-regulated GRP78 at both protein and mRNA levels(P<0.01),however,Hyperoside could reverse the expressions of CHOP and GRP78 in Palmitic acid-induced cells(P<0.01).Conclusion:Hyperoside can attenuate the endoplasmic reticulum stress and inhibit apoptosis of endothelial cells induced by Palmitic acid,raise the migration capacity.
