Experimental Study on Regulatory Effect of Buyang Huanwu Decoction(补阳还五汤)on Autophagy in Rats with Denervated Muscle Atrophy
Objective:To explore the regulatory effect of Buyang Huanwu Decoction on autophagy in dener-vated muscle atrophy rats,and to elucidate the protective mechanism of Yiqi Huoxue Therapy on denervated muscle atrophy.Methods:Models of nerve crush were prepared in rats,and the experiment was divided into sham operation group,model group,Mecobalamin group,Buyang Huanwu Decoction group,Buyang Huanwu Decoction+LY294002 group,with 8 rats in each group.Each group rats were given the corresponding medi-cine daily,on the 18th day,the wet weight ratio of the anterior tibialis muscle was calculated and morphological observation was performed.Western-blot was used to detect PI3K/PKB/mTOR(phosphatidylinositol 3-ki-nase/protein kinase/B/rapamycin target protein)signaling pathway proteins expressions.Results:The wet weight ratio of anterior tibial muscle of rats in the model group was significantly decreased(P<0.05),while it was significantly increased in the Buyang Huanwu Decoction group and Mecobalamin group(P<0.05),and it was decreased in Buyang Huanwu Decoction+LY294002 group.Morphological observation showed that the cross-sectional area of muscle fibers in the model group was decreased,the morphology was irregular,the structure of muscle fibers was disordered,connective tissue hyperplasia was increased and muscle fibers were sig-nificantly atrophic and degenerated.The proliferation of collagen fibers in Buyang Huanwu Decoction group was significantly reduced(P<0.05),and the morphology was regular.The muscle atrophy was aggravated and colla-gen fibers were increased significantly in the Buyang Huanwu Decoction+LY294002 group.The relative expres-sions of p-pi3k,p-PKB and mTOR proteins in Buyang Huanwu Decoction group and Mecobalamin group were significantly higher than those in the model group(P<0.05);the relative expression levels of ATG5 and becline-1 proteins were significantly decreased(P<0.05).The protein expressions of p-pi3k,p-PKB and mTOR were decreased in the Buyang Huanwu Decoction+LY294002 group,while the protein expressions of ATG5 and becline-1 were increased.Conclusion:Buyang Huanwu Decoction can play the protective role of denervat-ed muscle atrophy by inhibiting the excessive activation of autophagy-lysosome system through PI3K/PKB/mTOR pathway.
