目的:探讨熊果酸对H9C2心肌细胞H2O2过氧化损伤的保护作用及其与热休克蛋白(HSPs)的表达的相关性.方法:采用H2O2诱导H9C2心肌细胞氧化应激模型,实验分为正常对照组(Control)、H2O2组(H2O2)、熊果酸+H2O2组(Ursolic Acid,UA).CCK8检测各组细胞活力值,ELISA法检测培养基上清液 HSP27、HSP70、HSP90 含量,Western blot 检测各组细胞 HSP27、HSP70、HSP90、HSF1 蛋白表达水平.结果:与正常对照组相比,H2O2组细胞活力值降低,HSP27、HSP70、HSP90含量及HSP27、HSP70、HSP90、HSF1的表达水平上升(均P<0.05);与H2O2组相比,熊果酸+H2O2组细胞活力值升高,HSP27、HSP70、HSP9 0含量及HSP27、HSP70、HSP90、HSF1的表达水平上高(均P<0.05).结论:熊果酸可以通过促进HSPs的表达提高H9C2心肌细胞的抗氧化能力从而发挥对H2O2过氧化损伤的保护作用.
Research on Protective Effects of Ursolic Acid on H2O2-Induced Oxidative Damage in H9C2 Cardiomyocytes Based on Changes of Heat Shock Protein(HSP)Expression
Objective:To investigate the protective effects of Ursolic acid on H2O2-induced oxidative da-mage in H9C2 cardiomyocytes and its relationship with the expression of heat shock proteins(HSPs).Methods:H2O2 was used to induce oxidative stress in H9C2 cardiomyocytes,experiment were divided into the following groups:Control(normal control),H2O2(H2O2-treated),and Ursolic Acid+H2O2(Ursolic Acid,UA).Cell viability was determined by the CCK8 assay.The contents of HSP27,HSP70,and HSP90 in the cell culture su-pernatant were measured by ELISA.The protein expression levels of HSP27,HSP70,HSP90,and HSF1 in the cells were evaluated using Western blotting.Results:Compared to the control group,the cell viability in H2O2 group was decreased,HSP27,HSP70,HSP90 contents and HSP27,HSP70,HSP90,HSF1 protein expressions were upregulated(all P<0.05).Compared to the to the H2O2 group,the cell viability in UA group was in-creased,HSP27,HSP70,HSP90 contents and HSP27,HSP70,HSP90,HSF1 protein expressions were raised(all P<0.05).Conclusion:Ursolic acid can protect H9C2 cardiomyocytes against H2O2-induced oxidative damage by promoting the expression of HSPs and enhancing the antioxidant capacity.
