补肾活血方对去势骨质疏松小鼠骨-血管形成偶联调节作用的实验研究

目的:基于骨-血管形成偶联调节作用探讨补肾活血方对去势骨质疏松小鼠骨量及骨代谢的影响.方法:选取40只雌性ICR小鼠按随机数字表法分为假手术组(10只)和去势组(30只).去势组小鼠行卵巢去势手术后常规饲养8周,随机抽取3只小鼠证实骨质疏松造模成功后,按随机数字表法分为模型组和中药组,每组各10只.假手术组及模型组予以等量生理盐水灌胃.中药组成模后采用补肾活血方(8.6 mg/kg)灌胃干预,8周后处死小鼠收集样本.进行Micro-CT、TRAP染色、免疫荧光染色(EM-CN、CD31)、骨代谢指标检测.结果:Micro-CT显示模型组较假手术组骨小梁显著减少,中药组较模型组骨小梁增加.定量分析结果显示,模型组骨体积分数(BV/TV)、骨小梁数目(Tb.N)、骨小梁厚度(Tb.Th)低于假手术组,而中药组BV/TV、Tb.N、Tb.Th高于模型组,差异具有统计学意义(P<0.05).骨组织TRAP染色及定量分析显示,模型组TRAP阳性表达的数目高于假手术组,中药组阳性表达的数目少于模型组,差异具有统计学意义(P<0.05).模型组干骺端H血管的比例相比假手术组显著减少,中药组干骺端H血管的比例较模型组增加,差异具有统计学意义(P<0.05).与假手术组相比,模型组PINP水平降低,TRACP水平升高,差异具有统计学意义(P<0.05);中药组PINP水平较模型组升高,而TRACP降低,差异具有统计学意义(P<0.05).结论:补肾活血方可通过促进骨-血管形成,增加骨生成,减少骨吸收,提高去势骨质疏松小鼠的骨量.
Regulation Effect of Bushen Huoxue Formula(补肾活血方)on Osteogenesis and Angiogenesis Coupling in Ovariectomized Osteoporosis Mice
Objective:To explore the effect of Bushen Huoxue Formula on bone mass and bone metabolism in ovariectomized osteoporosis mice by regulating osteogenesis and angiogenesis Coupling.Methods:40 female ICR mice were randomly divided into sham group(10 mice)and ovariectomy group(30 mice)by random num-ber table method.The mice in the ovariectomized group were routinely raised for 8 weeks after undergoing ovari-ectomy surgery.3 mice were randomly selected to confirm successful osteoporosis modeling.Then,they were ran-domly divided into a model group and a Chinese medicine group,10 mice in each group.The sham operation group and model group were gavaged with equivalent saline.the Chinese medicine group were gavaged with Bushen Huoxue Formula(8.6 mg/kg).After 8 weeks of feeding,the samples were collected.Micro CT,TRAP staining,immunofluorescence staining(EMCN,CD31),and bone metabolism index were detected and analy-sised.Results:Micro-CT showed that the sham group was the most bone trabeculae,bone trabeculae of the model group was decrease,while that of the Chinese medicine group was increased compared to the model group.The quantitative analysis results showed that the bone volume fraction(BV/TV),number of trabeculae(Tb.N)and thickness of trabeculae(Tb.Th)in the sham group were higher than those in the model group and the Chinese medicine group,while the BV/TV,Tb.N,and Tb.Th in the Chinese medicine group were higher than those in the model group(P<0.05).Bone tissue TRAP staining and quantitative analysis showed that the number of TRAP positive expressions in the model group was higher than that in the sham group and the Chinese medicine group,while the number of TRAP positive expressions in the sham group was lower than that in the Chinese medicine group(P<0.05).The proportion of epiphyseal H-vessels in the model group was signifi-cantly reduced compared to the sham group,while the proportion of epiphyseal H-vessels in the Chinese medi-cine group was increased compared to the model group,but was lower than that of the model group(P<0.05).Compared with the sham group,the PINP level in the model group was decreased and the TRACP level was in-creased(P<0.05).The PINP level in the Chinese medicine group was increased compared to the model group,while TRACP was decreased,with a statistically significant difference(P<0.05).Conclusion:Bushen Huoxue Formula can promotes osteogenesis and angiogenesis,thereby increasing bone formation,reducing bone resorp-tion,and increasing bone mass in ovariectomized osteoporosis mice.
