Experimental Study on Anti-Liver Injury Effects of Silymarin Derivatives
Objective:To evaluate the anti-liver injury effects of Silymarin in vivo. Methods:48 C57BL/6 mice were randomly divided into 6 groups with 8 mice in each group as normal group,model group,23-substituted of Silybin derivative low dose group (25 mg/kg),medium dose group (50 mg/kg),high dose group (100 mg/kg) and Silybin control group (100 mg/kg). They were given relevant drugs (i. g. ) every day for 5 d. One hours after the final Ig medicine,APAP ( 350 mg/kg) was administrated intraperitoneally ( i. p. )to each group except normal group. Fasting without water and blood was collected from celiac vein 24 hours la-ter. Serological indexes of ALT,AST,MDA,GSH and GDH of hepatic tissue were determined,HE staining was used to observe the histopathological changes of liver tissue. Results:Different doses of the Silymarin derivatives could decrease the serological ALT and AST indexes,MDA and GDH levels in liver tissue of mice induced by paracetamol,and increase the content of GSH. At the same time,it can improve the liver tissue damage of paracetamol to different degrees,and show different degrees of anti-liver damage activities. Conclusion:Sily-marin derivatives can play a certain anti-liver injury effect by regulating MDA,GDH,GSH and other indexes in liver tissue.
