基于网络药理学探讨半夏白术天麻汤与羌活胜湿汤同病异治的生物学基础

目的 运用网络药理学分析半夏白术天麻汤与羌活胜湿汤的成分、靶点及机制差异,初步阐明"同病异治"在偏头痛中的内涵.方法 通过TCMSP检索半夏白术天麻汤和羌活胜湿汤的有效成分,并在SwissTargetPrediction中预测潜在靶点,通过TTD、GeneCards和OMIM数据库获取偏头痛的疾病基因,运用Cytoscape3.9.1软件进行数据分析,利用STRING数据库构建蛋白相互作用网络,用DAVID数据库进行通路富集分析.最后选取成分-靶点网络中度值较大的关键靶点和关键成分,使用AutoDock Vina软件进行分子对接.结果 半夏白术天麻汤及羌活胜湿汤共同靶点320个,半夏白术天麻汤特有靶点319个,羌活胜湿汤特有靶点66个.二者可能通过IL6、AKT1、TNF、VEGFA等共同关键靶点及HIF-1、IL-17等信号通路作用于偏头痛.其中,半夏白术天麻汤通过GABRB3、CHRM2、GABRB2等蛋白所富集的5-羟色胺能突触、神经活性配体-受体相互作用通路、MAPK信号通路发挥特有作用.羌活胜湿汤通过IL10、CAMK2B所在的PI3K-Akt等信号通路调节细胞存活等途径治疗偏头痛.分子对接结果显示,关键成分与关键靶点结合结构稳定.结论 半夏白术天麻汤主治内生风痰之头痛,羌活胜湿汤主治外感风湿之头痛.网络药理学分析发现二者既具有共同的靶点与作用通路,也有各自特有的靶点与作用通路,通过对差异靶点和通路的分析,可初步阐明同病异治不同证型头痛的生物学基础.
Discussion on the Scientific Connotation of Different Treatments for the Same Disease between Banxia Baizhu Tianma Decoction and Qianghuo Shengshi Decoction Based on Network Pharmacology
Objective To analyze the differences in composition,target and mechanism between Banxia Baizhu Tianma Decoction and Qianghuo Shengshi Decoction using network pharmacology;To preliminarily elucidate the connotation of"different treatments for the same disease"in migraine.Methods Retrieve the effective ingredients of Banxia Baizhu Tianma Decoction and Qianghuo Shengshi Decoction through the TCMSP database,and predict potential targets in SwissTargetPrediction,the disease genes of migraine were obtained by TTD,GeneCards,and OMIM database.The data were analyzed using the Cytoscape 3.9.1 software,and the STRING database was used to construct the PPI network.DAVID database was used for pathway enrichment analysis.Finally,the key targets and key components with large degree values in the component-target network were selected,and molecular docking was performed using AutoDock software.Results There were 320 common targets in Banxia Baizhu Tianma Decoction and Qianghuo Shengshi Decoction,319 targets specific to Banxia Baizhu Tianma Decoction,and 66 targets specific to Qianghuo Shengshi Decoction.They may act on migraine through IL6,AKT1,TNF,VEGFA and other common key targets,as well as HIF-1 and IL-17 signaling pathway.Among them,Banxia Baizhu Tianma Decoction exerted its specific effects through 5-hydroxytryptaminergic synapses enriched by proteins such as GABRB3,CHRM2,and GABRB2,the neuroactive ligand-receptor interaction pathway,and the MAPK signaling pathway.Qianghuo Shengshi Decoction could have unique therapeutic effects on migraine through the PI3K-Akt signaling pathway where IL10 and CAMK2B were located,and through the regulation of cell survival and other pathways.The results of molecular docking show that the key components bind to the key targets in a stable structure.Conclusion Banxia Baizhu Tianma Decoction is used to treat the migraine of endogenous wind and phlegm,while Qianghuo Shengshi Decoction is used to treat the migraine of exogenous wind and dampness.Network pharmacological analysis finds that both have common targets and pathways,as well as their own unique targets and pathways.By analyzing the differences in targets and pathways,the biological basis of the different types of migraine with different treatments for the same disease can be initially elucidated.
