基于网络药理学探讨清肾颗粒主要药物治疗慢性肾脏病作用机制

目的 基于网络药理学探讨清肾颗粒主要药物大黄、黄连、白花蛇舌草、丹参治疗慢性肾脏病(CKD)的作用机制.方法 利用TCMSP及SwissTargetPrediction数据库分别检索4味中药活性成分及其靶点,运用OMIM、GeneCards、TTD数据库获取疾病靶点,通过STRING数据库筛选核心靶点,并进行GO功能与KEGG通路富集分析,利用Cytoscape3.7.2软件构建蛋白相互作用网络.结果 共筛选出中药活性成分99种,药物靶点873个,活性成分治疗CKD交集靶点102个,主要活性成分包括槲皮素、木犀草素、丹参醇A等.核心靶点涉及VEGFA、EGFR、AKT1等.GO功能分析结果显示,VEGFA、EGFR、SRC等靶点参与CKD的治疗,KEGG通路富集分析结果显示,大黄、黄连、白花蛇舌草、丹参治疗CKD主要涉及HIF-1、PI3K-Akt、Rap1、MAPK等信号通路.结论 大黄、黄连、白花蛇舌草、丹参可能通过HIF-1、PI3K-Akt等信号通路,调控VEGFA、EGFR、SRC等靶点治疗CKD.
Exploration on the Mechanism of main drugs of Qingshen Granule in Treating Chronic Kidney Disease Based on Network Pharmacology
Objective To explore the mechanism of main drugs of Qingshen Granule of Rheum officinale,rhizoma coptidis,Hedyotis diffusa,and salvia miltiorrhizae in treating chronic kidney disease(CKD)based on network pharmacology.Methods TCMSP and SwissTargetPrediction databases were used to retrieve the active components and targets of of the four kinds of Chinese materia medica.OMIM,GeneCards and TTD databases were used to obtain disease targets.STRING database was used to screen core targets.GO function and KEGG pathway enrichment analysis was carried out.Cytoscape 3.7.2 software was used to construct PPI network.Results Totally 99 active components,873 action targets were obtained and 102 intersecting targets for the treatment of CKD with active components.The main active components included quercetin,luteolin,danshenol A,etc.The key targets involved VEGFA,EGFR,AKT1,etc.The GO function analysis results showed that targets such as VEGFA,EGFR,and SRC were involved in the treatment of CKD.The KEGG pathway enrichment analysis results showed that the treatment of CKD with Rheum officinale,rhizoma coptidis,Hedyotis diffusa and salvia miltiorrhizae mainly involved signaling pathways such as HIF-1,PI3K-Akt,Rap1,and MAPK.Conclusion Rheum officinale,rhizoma coptidis,Hedyotis diffusa and salvia miltiorrhizae may regulate target such as VEGFA,EGFR and SRC for CKD through signaling pathways such as HIF-1 and PI3K-Akt.
