摘要
目的 探索BCAT1在皮肤鳞状细胞癌(cutaneous squamous cell carcinoma,CSCC)表达及作用,并初步探索其可能的潜在机制.方法 利用免疫组织化学染色对人正常皮肤组织及CSCC组织中BCAT1 的表达进行检测.利用转染技术对敲低人皮肤鳞癌SCL-1 细胞中BCAT1 表达后,检测细胞活力、增殖水平、迁移及侵袭能力;Western blot检测细胞增殖和侵袭相关的关键蛋白(PCNA、Snail、N-cad、Twist、MMP-7)的表达水平.结果 与正常皮肤组织相比,CSCC组织中BCAT1的表达水平显著增加;与NC-shRNA组相比,BCAT1-shRNA组细胞活力、增殖水平、迁移及侵袭能力均显著降低;与NC-shRNA组相比,BCAT1-shRNA组细胞中增殖和侵袭相关蛋白(PCNA、Snail、N-cad、Twist、MMP-7)表达水平均显著降低.结论 BCAT1在CSCC细胞中表达上调,且能促进CSCC细胞增殖、迁移和侵袭.
Abstract
Objective To explore the expression and function of BCAT1 in cutaneous squamous cell carcinoma(CSCC)and to preliminarily investigate its potential underlying mechanisms.Methods Immunohistochemistry staining was used to detect the ex-pression of BCAT1 in normal human skin tissues and CSCC tissues.Following the knockdown of BCAT1 expression in human skin squamous cancer SCL-1 cells using transfection techniques,cell viability,proliferation levels,migration,and invasion capabilities were examined;Western blot analysis was utilized to measure the expression levels of key proteins related to cell proliferation and invasion(PCNA,Snail,N-cad,Twist,MMP-7)in the cells.Results Compared to normal skin tissues,the expression level of BCAT1 in CSCC tissues significantly increased.Compared to the NC-shRNA group,cell viability,proliferation levels,migration,and invasion capabili-ties were significantly reduced in the BCAT1-shRNA group.Additionally,compared to the NC-shRNA group,the expression levels of proteins related to proliferation and invasion(PCNA,Snail,N-cad,Twist,MMP-7)in the BCAT1-shRNA group cells were significantly decreased.Conclusion BCAT1 is up-regulated in CSCC cells and can promote proliferation,migration,and invasion of CSCC cells.
