摘要

炎症作为一把"双刃剑",与恶性肿瘤的发生、发展及对靶向、免疫治疗的疗效关系密切,可促进或者抑制肿瘤的进展.其中,IL-6作为炎症细胞因子中的重要成员,调控多种肿瘤的病理过程,在临床中被广泛应用并监测,可作为肿瘤细胞增殖、浸润的生物标志物,被认为是衔接炎症与肿瘤最为核心的细胞因子之一.近年来靶向、免疫治疗的出现显示出了良好的疗效,但靶向治疗后部分患者易产生耐药,免疫治疗在实体瘤患者中的平均有效率也较为有限,如何提高靶向和免疫治疗疗效,扩大受益人群是亟待解决的问题.抑制IL-6 及其信号转导通路可重启肿瘤-免疫循环,打破免疫抑制的掣肘同时改善免疫微环境,使靶向、免疫治疗更为行之有效.

Abstract

Inflammation acts as a"double-edged sword",closely related to the occurrence,development,and effectiveness of targeted and immunotherapy treatments in malignant tumors,capable of either promoting or inhibiting tumor progression.Among them,IL-6,as an important member of inflammatory cytokines,regulates the pathological processes of various tumors.It is widely used and monitored in clinical settings and can serve as a biomarker for tumor cell proliferation and infiltration,considered one of the core cytokines linking inflammation and cancer.In recent years,the emergence of targeted and immunotherapy has shown promising effectiveness,but some patients develop resistance to targeted therapies,and the average effectiveness of immunotherapy in solid tumor patients remains limited.How to enhance the effectiveness of targeted and immunotherapy and expand the beneficiary population is an urgent problem to be solved.Inhibiting IL-6 and its signaling pathway can restart the tumor-immune cycle,breaking the constraints of immune suppression while improving the immune microenvironment,making targeted and immunotherapy more effective.
