Effect of CeNPs for DOX Induced Cardiotoxicity and Their Influence on SOD
Objective To investigate effect of CeNPs(nano ceria)for DOX(doxorubicin)induced myocardial cell injury,and influence of CeNPs on SOD activity in DOX induced myocardial cell injury.Methods The experiment established injury model with H9C2 rat cardiomyocytes after DOX treatment,and randomly divided cells into 5 groups:blank control group,DOX injury group,CeNPs group,DOX+CeNPs group,and DOX+DEX(positive protective drug)group for experiment,and measured survival rate by CCK-8 method,protein expression by Western Blot method,and oxidative stress indicator SOD activity by biochemical method.Results CCK-8 method showed cell survival rate in DOX+CeNPs group was significantly higher than DOX group(P<0.0001).Western Blot methods showed,expression level of Bax protein in DOX+CeNPs was lower than DOX group,(P<0.0001),expression level of Bcl-2 protein was higher,(P<0.05).Biochemical detection:DOX+CeNPs group showed higher SOD activity than DOX group,(P<0.05).Conclusion(1)CeNPs can improve cell survival rate after DOX treatment;(2)CeNPs can alleviate cell apoptosis after DOX treatment by regulating expression of apoptosis related proteins;(3)CeNPs can improve cellular oxidative stress imbalance after DOX treatment by increasing SOD activity.
