Expression and Clinical Correlation Analysis of PD-1/PD-L1 Combined with CD8+TILs in Cervical Cancer Tissue
Objective To explore expression and clinical correlation of CD8+TILs and PD-1/PD-L1 molecules in cervical cancer tissues.Methods The paper chose 152 cervical cancer patients admitted to Bengbu Medical College the First Affiliated Hospital from January 2016 to January 2020 as study subjects,and collected basic patient information and measured PD-1/PD-L1 positivity and CD8+TILs density with immunohistochemical staining.152 cases were divided them into group A(n=40),group B(n=59),group C(n=43),and group D(n=10)according to PD-1/PD-L1 expression and CD8+TILs density.Relationship between PD-1/PD-L1,PD-L1/CD8+Til density and clinical pathological characteristics of cervical cancer,as well as correlation were analyzed between CD8+TILs and PD-1/PD-L1.Survival status of cervical cancer patients with different PD-1/PD-L1 expression and CD8+TILs density was analyzed.Results Ppositive expression rates of PD-L1 and PD-1 were 65.13%and 61.18%respectively;positive expression of PD-L1 was closely related to tumor size,depth of interstitial infiltration,FIGO staging,lymph node metastasis,and parametrial infiltration(P<0.05).Positive expression of PD-1 was not closely related to clinical and pathological characteristics of cervical cancer(P>0.05).High-density and low-density cases of CD8+TILs were 82 and 70 respectively.Spearman test analysis results showed expression of PD-1 and PD-L1 was positively correlated(r=0.742,P<0.001)and negatively correlated(r=-0.385,P<0.001)with density of CD8+TILs respectively.Four groups showed significant difference in tumor size,FIGO staging,and lymphatic vessel invasion(P<0.05).FIGO staging,degree of differentiation,lymphatic vessel invasion,and lymph node metastasis was independent factors affecting OS in cervical squamous cell carcinoma patients.Survival time of PD-L1 positive group was shorter than PD-L1 negative group(according to the Log Rank test,χ2=20.986,P=0.000<0.05).Survival time of CD8+TILs low-density group was shorter than CD8+TILs high-density group(according to the Log Rank test,χ2=40.788,P=0.000<0.05).Survival time of D-L1-/CD8+TILs high-density group was higher than the other three groups(according to the Log Rank test,χ2=47.145,P=0.000<0.05).Conclusion PD-L1 is highly expressed in cervical cancer,and PD-1 and PD-L1 expression is positively and negatively correlated with CD8+TILs density respectively.Combination of PD-1/PD-L1 and CD8+TILs has high clinical application value in prognosis evaluation of cervical cancer.
