Effect Research of LncRNA HOTAIR and miRNA in the Pathogenesis of Bladder Cancer
Objective To study the effect of LncRNA HOTAIR and miRNA in the pathogenesis of bladder cancer. Methods The paper collected study subjects from five hospitals,including Mudanjiang City the First and Second People's Hospital,the Red Flag Hospital and the Second Affiliated Hospital of Mudanjiang Medical College,and Mudanjiang Cancer Hospital,and chose 200 bladder cancer patients as the case group and 200 healthy volunteers as the control group from February 2020 to February 2023. The expression of LncRNA HOTAIR in plasma and tissue of bladder cancer patients and healthy people was detected by real-time quantitative PCR to explore correlation between HOTAIR and the clinicopathological characteristics of bladder cancer. Correlation was explored between LncRNA HOTAIR and candidate miRNAs,to elaborate the LncRNA HOTAIR miRNA regulatory network. Results The expression Ct value of LncRNA HOTAIR protein in the case group was lower than the control group (P<0.05). The expression of LncRNA HOTAIR of patients with bladder cancer tumor size ≥ 4 cm was lower than the control group (P<0.05). The expression of LncRNA HOTAIR of patients with clinical stage Ⅱ was lower than the control group (P<0.05). The expression of LncRNA HOTAIR of highly differentiated patients was lower than the control group (P<0.05). The expression of LncRNA HOTAIR of patients with infiltration depth ≥ 1/2 was lower than the control group (P<0.05). The expression of LncRNA HOTAIR of patients with lymph node metastasis was lower than the control group (P<0.05). The LncRNA HOTAIR of the case group patients showed significant negative correlation with miR-34a-5p,miR-214-5p,and miR-218-5p (P<0.05). There was significant positive correlation between LncRNA HOTAIR and miR-218-5p in the control group,(P<0.05). Conclusion LncRNA HOTAIR and miRNA are involved in the regulation of cancer-related genes.
