Experimental Study on Correlation between Polycystic Ovary Syndrome and Psychological Stress
Objective The polycystic ovary syndrome (PCOS) model was established by DHEA exposure combined with chronic unpredictable stress (CUMS) in female rats to study effect of adverse psychological stress in the occurrence and development of PCOS,and investigate effect of DHEA exposure combined with CUMS on insulin resistance,inflammatory factors,sex hormones and body mass index of PCOS in rats.Methods The paper chose 48 female SD rats,and divided them into blank control group randomly,DHEA group,psychological stress group and combined group (DHEA combined psychological stress group).The blank group was treated with standard feed,the PCOS model group was injected with DHEA subcutaneously (6mg/100g),and the chronic stress group was modeled with CUMS method,and a stimulus was randomly given each day to avoid the method repetition during two consecutive days and the sequential repetition during two consecutive weeks.The PCOS combined with chronic stress group was treated with the above two modeling methods for 28 days.After 28 days,the height and weight of four groups of rats was measured,and T,FSH,LH,FINS and inflammatory factors (IL-1 and TNF-α) in serum of rats were detected by ELISA.FPG and blood glucose were detected by blood glucose analyzer.Ovarian tissue morphology was observed,body mass index (BMI) and insulin resistance index (HOMA-IR) were calculated to evaluate insulin resistance.Results PCOS rat model was established successfully.Compared with blank group,serum LH and T contents in model group were significantly higher,(P<0.05).LH/FSH ratio was significantly higher,(P<0.05),and FSH contents were significantly lower,(P<0.05).Conclusion Psychological stress plays an important role in the occurrence and development of PCOS.
