Objective To investigate whether the osteogenic differentiation ability and biological characteristics of rJBMMSCs derived from diabetic osteoporosis have changed.Methods Diabetic osteoporosis(DOP)model was created in GK rats.After successful modeling,cells were extracted from mandibular bone of rats,and identified.Wistar rats were used as control group.Cell proliferation ability was detected by CCK8 assay,cell clonogenesis ability was detected by plate cloning assay,and cell migration ability was detected by scratch assay.The osteogenic differentiation ability was detected by ALP staining,ARS staining,qRT-PCR and Western blot.Results DOP rJBMMSCs showed decreased cell proliferation,clonogenesis and cell migration compared with Control group.The staining area of ALP in DOP group was smaller and lighter than that in Control group,and the activity of ALP was significantly decreased.ARS staining showed that there were less mineralized nodules in DOP group than in Control group.The results of qRT-PCR and Western blot showed that the mRNA and protein levels of osteoblast-related factors in DOP group were significantly decreased.Conclusion Compared with the control group,the biological characteristics of DOP rJBMMSCs were changed,and their osteogenic differentiation ability was significantly impaired.
