Objective This study aims to analyze the expression levels and clinical significance of Integrin Subunit Alpha 3 (ITGA3) in Head and Neck Squamous Cell Carcinoma (HNSCC) using online databases. Methods TCGA and CPTAC oncology gene databases were used to analyze the relationship between ITGA3 gene,protein,and promoter methylation levels in HNSCC and various tumor characteristics such as age,gender,race,smoking status,clinical stage,lymph node infiltration,HPV infection,and TP53 mutations. Additionally,the relationship between ITGA3 expression in HNSCC patient tissues and overall survival was analyzed using the UALCAN website. Results Compared with normal controls,ITGA3 expression was increased and ITGA3 promoter methylation was decreased in HNSCC patients (P<0.001). The expression levels of ITGA3 gene and protein were correlated with patient gender,HPV infection,TP53 mutations,tumor grading,HIPPO pathway alterations,and smoking status. Higher ITGA3 expression levels and female patients showed poorer prognoses. The String database revealed interactions between ITGA3 and 10 genes,primarily involved in cell adhesion and hemidesmosome assembly. Moreover,the TIMER database displayed a positive correlation between ITGA3 levels and specific immune cells. Conclusion ITGA3 functions as an oncogenic factor in HNSCC,with its expression associated with gender,tumor grading,immune cell infiltration,and smoking status. It holds potential as a novel therapeutic target and prognostic marker for HNSCC treatment and prognosis.
